ObjectiveTo investigate the influencing of curcumin on expression of COX-2inhuman hepatocellular carcinoma BEL-7402cells.Methods1.Various concentrations of curcumin(0μmol/L、2.5μmol/L、5μmol/L、10μmol/L、15μmol/L、20μmol/L) treated human hepatocellular carcinomaBEL-7402cells. The cell proliferation were assayed by CCK-8method. Theexpressions of COX-2in human hepatocellular carcinoma BEL-7402cellswere checked by RT-PCR and Western Blot.2. human hepatocellularcarcinoma BEL-7402cells were treatment with NS-398200μmol/L plus10μmol/L of curcumin and alone treated with10μmol/L of curcumin, theexpressions of VEGF mRNA、protein were checked by RT-PCR and WesternBlot.Results1.The cell proliferation rate were significant difference between controland different concentrations of curcumin (P<0.01).2. Curcumin candown-regulate the expression of COX-2mRNA with the increase ofconcentration gradually and the difference is significant compared with thecontrol(P <0.01);3. Curcumin can down-regulate the expression of COX-2protein with the increase of concentration gradually and the difference issignificant compared with the contro(lP <0.01).4. The expressions of VEGFmRNA、VEGF protein in10μmol/L of curcumin group,200μmol/L of NS-398 plus10μmol/L of curcumin group were respectively reduced vs control, and thedifference is significant compared with the control(P <0.01), The expressionsof VEGF mRNA、VEGF protein in200μmol/L of NS-398plus10μmol/L ofcurcumin were respectively reduced vs10μmol/L of curcumin group(P <0.01).Conclusion1.Inhibitory effect of curcumin on expression of COX-2mRNA and proteinin human hepatocellular carcinoma BEL-7402cells;2. Curcumin inhibitingtumor angiogenesis in human hepatocellular carcinoma BEL-7402cells maythrough the way of COX-2/PGE2/VEGF.
