Purpose:This study is to explore the effect of curcumin on TLR4-mediated of MyD88+ovarian cancer cells secrete IL-6and VEGF, and the role of Curcumin to increase the sensitivity of ovarian cancer cells to paclitaxel.Method: we measured induced by paclitaxel in human ovarian cancer SKOV-3cells and A2780cells to secrete IL-6and VEGF in concentration by ELISA, and the concentration of LPS, curcumin, and different concentrations of curcumin combined with paclitaxel or LPS-induced. We were used to detect the growth inhibitory effect of paclitaxel on human ovarian cancer SKOV-3cells and A2780cells, and the effect of curcumin and LPS by MTT.Result:(1) paclitaxel and LPS are dose-dependent manner to promote SKOV-3cells secrete IL-6(P<0.05), but no effect on the secretion of VEGF (P>0.05). Curcumin are dose-dependent inhibition of SKOV-3cells to secrete IL-6and VEGF (P<0.05), and unhandled A2780cells secrete very small amounts of IL-6. Different concentrations of taxol, LPS or curcumin treatment, no significant changes in IL-6concentration (P>0.05). A2780cells secrete high levels of VEGF. Different concentrations of taxol, LPS or curcumin treatment, no significant changes in the concentration of VEGF (P>0.05).(2) Paclitaxel dose-dependent manner inhibited the survival of SKOV-3cells and A2780cells, and their IC50were (0.037±0.005) μmol/L and (0.029±0.003) μmol/L. LPS can reduce SKOV-3cells to paclitaxel sensitivity (P<0.05),its IC50was (0.060±0.005) umol/L.LPS does not affect A2780cells to paclitaxel sensitivity (P>0.05), its IC50was (0.030±0.004) μmol/L. Curcumin can increase the SKOV-3and A2780cells to paclitaxel sensitivity (P<0.05), and their IC50were (0.021±0.004) μmol/L and (0.016±0.002) μmol/L.Conclusion: Experimental results show that MyD88+human ovarian cancer cells themselves can secrete IL-6, and curcumin in a dose dependent manner antagonized paclitaxel and LPS-induced of MyD88+human ovarian cancer cells to secrete IL-6, and this effect depends on the existence of TLR-4/of MyD88+signaling pathway.MyD88+and MyD88-human ovarian cancer cells themselves are the high expression of VEGF, but taxol and LPS does not promote the expression of VEGF. Curcumin dose-dependent manner antagonist of MyD88+human ovarian cancer cells secrete VEGF, While no effect of curcumin of MyD88-human ovarian carcinoma cell.Curcumin can increase paclitaxel on ovarian cancer cell growth inhibition, this role is not only dependent of TLR-4/of MyD88+signaling pathway, there may be other ways.
