| ObjectiveBy detecting and analyzing the expressions of gene associated withretinoid-interferon mortality-19(GRIM-19) and its target gene product signaltransducer and activator of transcription3(STAT3) in pancreatic carcinomatissues and pancreatic tumor tissues of human, to research the role of GRIM-19,STAT3in the occurrence and development of pancreatic carcinoma.MethodsThe mRNA expressions of GRIM-19and STAT3in30cases of pancreaticcarcinoma tissues and in22cases of pancreatic tumor tissues were detected byRT-PCR. Applying immunohistochemistry (IHC) dyeing and western blotting todetect the protein expressions of GRIM-19and STAT3in30cases ofpancreatic carcinoma and in22cases of pancreatic tumor tissues. Thencompare the difference of their expressions, analyze the correlation of theirexpressions. And analyze statistically the relationship of their expressions withthe relevant features of clinical pathology of pancreatic carcinoma.ResultsExpressions of GRIM-19and STAT3were observed in both pancreaticcarcinoma tissues and pancreatic tumor tissues. The mRNA and protein levelsof GRIM-19in pancreatic carcinoma tissues were lower than that in pancreatic tumor tissues (P<0.01), and the expression of GRIM-19was associated with thedifferentiation degree of pancreatic carcinoma (P<0.05). By comparison, themRNA and protein levels of STAT3in pancreatic carcinoma tissues were higherthan that in pancreatic tumor tissues (P<0.01), and the expression of STAT3was associated with the differentiation degree and lymph node metastasis ofpancreatic carcinoma (P<0.05). Their expressions had nothing to do with thegender and age of patients (P>0.05). There was significantly negativecorrelation between the expressions of GRIM-19and STAT3in pancreaticcarcinoma tissues (r=-0.494, P<0.01).ConclusionsThere were expressions of GRIM-19and STAT3in pancreatic carcinomatissues and pancreatic tumor tissues. The low expression of GRIM-19and thehigh expression of STAT3existed in pancreatic carcinoma, and there wassignificantly negative correlation between the expressions of GRIM-19andSTAT3in pancreatic carcinoma. The different expressions may be connectedwith the growth and development of pancreatic carcinoma. |
