Objective:To study Mucin5AC(MUC5AC) and interleukin 13 (IL-13) expression and relativity in mouce lungs of COPD.to provide an new therapeutic target for COPD.Methods:Twenty male wistar mice were randomly divided into control group and COPD group, eight mice control group,twelve mice COPD group.The lung tissue was obtained and pathological analysis was done through Hematoxylin eosin (HE), MUC5AC and IL-13 expression were detected by immunohistochemical SP method,respectively.Results:1.The pathological morphology sbowed that airway epithelia were intact,cILiary arrangements were regular and the structures of alvcoli were normal in control group.COPD group:hyperemia and edema of mucous membrances,mucinous secretious of casts filling airways,hypertrophy of mucous glands and goblets cells,squamous metaplasia or dysplasia with loss of cilia of bronchial epithelium. Bronchia/bronchiolar mucous plugging inflammation and fibrosis,infiltrating by mononuclear cells and other inflammation cells. As the lumen of the bronchus becomes narrowed increased airways resistance is a feature of the advance cases and cause severe dyspnoea and limitation of activity. With diffuse forms,lungs can becomes voluminous and pillowy. microscopically,air spaces are enlarged;Walls thinned and septal capillaries compressed and bloodless and rupture of walls,with advanced diseases,adjacent alveoli become confluent,creating large airspace,may produce honeycombing.2.The expression of the MUC5AC and IL-13:Both Muc5ac and IL-13 positive products were located in the cytoplasm of airway epithelial cells. The expression of the MUC5AC and IL-13 in lung of group COPD was very significantly higher than that of group control. The expression of MUC5AC had posive collation with that of IL-13 (r=0.60,P<0.05)Conclusion:1. The model of COPD is established by passive smoking cigarettes.2. The expression of the MUC5AC and IL-13 in lung increased significantly,which explain that its play important roles in the invasive process of COPD.Mucus hypersecresion is the major pathological feacture of COPD.3. IL-13 is a critical cytokine inducing airway mucus overproduction,and may increase MUC5AC expression.
