Objective:Establish human gastric cancer in nude mice NUGC-4orthotopic xenograft model, to discover the mechanism of ginsenoside Rg3combined5-Fu restraining the metastasis of the human gastric, the influence of the expression of CD133in gastric cancer tissue,and discover the effect of ginsenoside Rg3on hematopoietic.Methods:1.By using the method of in situ transplantation, Establish people gastric cancer NUGC-4tumor strains in BALB/C nude mice back subcutaneous, and observe survival after transplantation tumor mice.2.Completely random methods32only nude mouse model can be divided into four groups, namely:the physiological saline group ginseng saponin Rg3group,5-Fu group and ginseng saponin Rg3+5-Fu group, each group of eight, after orthotopic transplantation9days, tail intravenous saline group was given saline water, each only0.2mL/second, Q2d x15;5-Fu group was given intraperitoneal injection of5-Fu injection, according to the standard20mg/kg,1times a week,3weeks in a row, Rg3group was given ginsenosides Rg3tail intravenous injection, according to the1mg/kg standard,Q2d x15;Rg3ginsenosides+5-Fu group was given intraperitoneal injection of5-Fu injection, according to the standard20mg/kg,1times a week,3weeks in a row, at the same time, according to standard according to1mg/kg, ginsenosides Rg3Q2d x15.3. Determination of tumor weight,and calculat the Inhibition rate,the inhibition rate of Ginsenoside Rg3group,5-Fu group,Rg3+5-Fu group were48.6%,69.2%,78.5%, the weight of xenografts of ginsenoside Rg3group,5-Fu group,Rg3+5-Fu group,were Lower significantly than the saline group,the difference was statistically significant(P<0.05).3.Calculated the distant metastasis of each group. Compared with the saline group,the distant metastasis rate of ginsenoside Rg3group,5-Fu group,Rg3+5-Fu group was significantly lower,the difference was statistically significant (P<0.05);The Rg3+5-Fu group has the lowest rate of distant metastasis.the distant metastasis rate of ginsenoside Rg3group and5-Fu group,also was lower than the saline group.the difference was statistically significant (P<0.05).4.CD133-positive rate of tumor tissues in each group were as follows:saline group86.2%, Rg3group36.7%,5-Fu group38.6%, Rg3+5-Fu group26.5%, compared with the saline group, the other three groups can inhibited the expression of CD133in tumor tissue, the difference was statistically significant (P<0.05), especially Rg3+5-Fu group, it has a significant statistically difference with the saline group.5. Blood smear showed:5-Fu group of white blood cells was significantly reduced, ranging from the size of mature red blood cells, platelets less aggregation. There are a larger number of white blood cells, red blood cell morphology was normal mature platelets piles visible in Rg3group and saline group, there was a significant difference compared with the5-Fu group, but the difference between saline and Rg3group of small, non-statistically significant. Rg3+5-Fu group than in the5-Fu group the number of white blood cells slightly, red blood cell morphology is also more rules, platelet aggregation is also relatively high, the difference was statistically significant.5-Fu on mouse hematopoietic function has some damage, but joint the ginsenosides Rg3can rebound after hematopoietic function in mice, indicating that ginsenoside Rg3on chemotherapy mouse hematopoietic function has a protective effect.Conclusion:l.Nude mouse orthotopic transplantation models was established successfully by using the gastric cancer NUGC-4tumor strains.2.Ginseng saponin Rg3joint the5-Fu can suppress the orthotopic transplantation tumor growth of nude mice significantly,at the same time,it can reduce the distant metastasis rate. Ginseng saponin Rg3joint the5-Fu can reduce the expression of CD133in tumor tissue. And tumor tissues and tumor distant metastasis of CD133positive correlation.3.Ginsenoside Rg3on chemotherapy mouse hematopoietic function has a protective effect.
