PART ONE:EFFECT OF COMPOUND GLYCYRRHIZINON TH17/TREG CELLS IN CHILDREN OF HENOCH-SCHONLEIN PURPURAObjective: To investigate the role of Th17、 Treg cells in thepathogenesis of childhood Henoch–Schonlein purpura(HSP), and furtherexplore the clinical effectiveness and immunomodulatory effects ofCompound Glycyrrhizin(GL) in HSP.Methods:27HSP patients were chose as GL-treated group, another26patients were used as conventional therapy group. Besides0.5-2ml/kgGL was injected in GL-treated group, other treatments were same.25ageand sex-matched healthy children were used as healthy controls. Bloodsamples of patients were obtained at the acute stage and after7daystreatment. Using intracellular staining, frequencies of peripheral Th17andTreg cells were detected by flow cytometry.Results:1.Clinical effectiveness: Renal complications in GL-treated group were relieved and recurrence of HSP within1month was lower.2.Frequency of Treg and Th17cell: Compared with healthy controls, thefrequency of Treg in GL-treated group was significantly down-regulatedduring acute phase(P=0.010); and it was up-regulated aftertreatment(P=0.059). As for conventional therapy group, the frequency ofTreg was lower in post-treatment compared with pre-treatment and healthylevel(P=0.006, P=0.013). The frequency of Th17in acute phase ofGL-treated group was significantly higher than post-treatment and healthylevels(P=0.024,P=0.013); The trend in conventional therapy group was thesame as GL-treated group(P=0.014), but neither one was different fromhealthy level(P=0.102,P=0.235).Conclusion:1.GL could relieve the renal complications and decreaserecurrence of HSP in a short period of time.2.Treg and Th17might in partcontribute to the process of HSP.3.GL could help to up-regulate Treg anddown-regulate Th17level, thus regulating the Th17/Treg deviation, andalleviating cellular immunity in HSP. PART TWO:EFFECT OF COMPOUND GLYCYRRHIZINON SERUM IL-17、IFN-γ、TNF-α、IP-10IN CHILDREN OFHENOCH-SCHONLEIN PURPURAObjective:To investigate the role of serum IL-17、IFN-γ、TNF-α、IP-10in the pathogenesis of childhood Henoch–Schonlein purpura(HSP),and further explore immunomodulatory effects of CompoundGlycyrrhizin(GL) in HSP.Methods:27HSP patients were chose as GL-treated group, another26patients were used as conventional therapy group. Besides0.5-2ml/kgGL was injected in GL-treated group, other treatments were same.25ageand sex-matched healthy children were used as healthy controls. Bloodsamples of patients were obtained at the acute stage and after7daystreatment. Serum IL-17、IFN-γ、TNF-α、IP-10level were detected byELISA.Results:1.Serum IL-17、IFN-γ level in GL-treated group were higherthan post-treatment and healthy levels (P=0.021,P=0.013; P=0.021,P=0.010), and they were down-regulated after treatment (P=0.052,P=0.154). As for conventional therapy group, no difference was detectedbetween pre-treatment and post-treatment (P=0.218,P=0.970).2.SerumTNF-α、 IP-10level in GL-treated group were down-regulated aftertreatment (P=0.011,P=0.037). But no difference was detected betweenpre-treatment and post-treatment in conventional therapy group (P=0.247,P=0.709). Conclusion:GL could help to down-regulate serum IL-17、IFN-γ、TNF-α、IP-10level, thus regulating cytokine imblance, and alleviatingcellular immunity in HSP.
