2-Naphthol is an organic synthesis of raw materials and intermediates,and rubber antioxidant, dressing agent, bactericide, preservatives,preservatives, prevention and control of parasitic and anthelmintic drugsetc. Recently, some unscrupulous traders have2-Naphthol beyond thescope of application in vegetables, long time use will cause certain damageto the human body.2-Naphthol has strong corrosive, strong stimulatingeffect on the skin. Easily absorbed through the skin, are toxic to thecirculation of the blood and kidney. In addition, it can cause cornealinjury.In recent years,2-Naphthol as preservatives are widely used in fruit.GB2760"hygiene standards for use of food additives" provisions of thescope of use of2-Naphthol as the surface treatment of fresh fruit (onlycitrus), the maximum amount of0.1g/kg, the residues of mg/kg≤70.China only allows the use of Macao area, but has not enacted residuelimits.This research has established a sensitive, accurate, simple, safe,rapid, simultaneous determination of fruit and vegetables, HPLC-FLD andHPLC-MS/MS detection methods, the paper consists of two parts. Part I Determination of2-Naphthol in fruits andvegetables by HPLC-FLD0bjective: To develop an HPLC-FLD method for the determinationof2-Naphthol in fruits and vegetables.Methods: The analytes were separated on a Pickering C18(250mm×4.6mm,5μm), Mobile phase: acetonitrile and water,(50+50, v/v);Column temperature:30℃; The detection wavelength: λex=250nm,λem=410nm;Injection volume:20L; Detector:Fluorescence detector.Results:2-Naphthol had good linear relationships in the range of0.50mg/L~50.00mg/L, the correlation coefficient was higher than0.9999;Average recoveries of2-Naphthol at the concentration of0.05,0.10,0.50mg/kg ranged was83.3%~101.2%and relative standard deviation rangewas4.3%~9.3%.Conclusions: the extracted with acetonitrile, purified amino solidphase extraction column, separation of liquid chromatography,fluorescence detection, To establish a rapid detection method of2-Naphthol in fruits and vegetables, the method is simple, rapid, sensitivewith good accuracy and precision.Part II Determination of2-Naphthol in fruits andvegetables by HPLC-MS/MS0bjective: To develop HPLC-MS/MS method for the determinationof2-Naphthol in fruits and vegetables. Methods: The analytes were separated on an Shimadzu VP-ODS C18(150mm2.0mm,5m); Mobile phase: acetonitrile2mmol/Lammonium acetic; Gradient elution; Flow rate:0.30mL/min; Columntemperature:35℃; Injection volume:30.00ul.MS/MS reference conditions: Detection methods: MRM; CUR:45psi; GS1:30psi; GS2:20psi; CAD:9psi; IS:-4500V; TEM:600C.Results: There was good linearity in the range of50.00μg/L-1000.00μg/L (r2≥0.9997). The limit of detection of the method was10μg/kg. Therecovery range was from70.4%to105.5%at the spiked level of10μg/kg,20μg/kg and50μg/kg in deferent samples, and relative standard deviationrange was from4.1%to10.6%.Conclusions: This method is of high accuracy, high sensitivity, goodrepeatability, detection and confirmation for2-Naphthol in fruits andvegetables.
