Objective To explor if2-methoxyestradiol combined withtraditional chemotherapy cyclophosphamide can improve curative effectby establishing an advanced breast cancer model of BALB/c4T1mice.Method Establishing advanced breast cancer model ofBALB/c4T1mice, which were randomly divided into4groups, called thecontrol group,2-ME2group, CTX group and2-ME2combined with CTXgroup, then we observed tumor growth situation. After3weeksContinuous drug intervention, the mice were killed at the end of theexperiment. Then the implantation tumors and lungs of the mice weretaken out and inhibition tumors rate and pulmonary metastasis rate wascalculated. Detected HIF-1α and VEGF protein expression andmicrovascular density (MVD) of the implantation tumors byimmunohistochemistry.Results1、Inhibition tumors rate: the combination group>2-ME2group or CTX group (P<0.05).2、pulmonary metastasis rate:2-ME2group was lower than the control group, the combination groupwas significantly lower than2-ME2group or CTX group. there werestatistically significant (P<0.05).3、the combination group MVD andHIF-1α, VEGF protein was significantly lower than the2-ME2group or CTX group (p <0.05).Conclusion1、Which2-ME2combined with CTX is bettercurative effect than only CTX or2-ME2to advanced breast cancer modelof BALB/c4T1mice.2、Co-treated with2-ME2and CTX have synergy to inhibiting tumorangiogenesis.
