OBJECTIVE: In immortalized nasopharyngeal epithelial NP69cells of knockdownSTGC3gene expression, screenned and identified83differentially expressed proteinmolecules utilized proteomics technology. this study choose one overexpressionprotein, membrane protein mucin3(ANXA3), one low expression proteinnicotinamide N-methyltransferase (NNMT), to verificate and analyze the significanceof ANXA3and NNMT protein expression in NPC cells and tissues, and explore therelationship between NNMT, ANXA3and STGC3gene expression, provideexperimental basis for clarifing the role of STGC3gene in nasopharyngealdevelopment.METHODS: RT-PCR and Western-blot technique to detect expression of ANXA3and NNMT in NP69cell lines that knockdown STGC3expression, nasopharyngealcarcinoma cell lines CNE1and CNE2and nasopharyngeal carcinoma tissues;immunohistochemical staining detect ANXA3and NNMT expression innasopharyngeal carcinoma, and analyze the relationship and significance betweenANXA3, NNMT and STGC3.RESULTS:1.ANXA3mRNA expression levels were higher in NP69cells of low expressionSTGC3, nasopharyngeal cells CNE1and CNE2, and nasopharyngeal carcinoma tisses,however the NNMT gene expression is reduced, compared with the controlgroup(NP69Cell, unnasopharyngeal carcinoma tisses) respectively, the differencewas statistically significant (P <0.05).2.The protein results were consistent with the gene test results: low expressionSTGC3NP69cells, nasopharyngeal CNE1and CNE2cells, nasopharyngealcarcinoma tissues are present ANXA3protein higher expression compared with thecontrol group, the difference was statistically significant (P <0.05). 3.Immunohistochemical staining results display: ANXA3protein localized in thecytoplasm and cell membrane, NNMT protein expressed at cytoplasm and nucleus,mainly the cytoplasm, the positive stainning rate of ANXA3protein was83.3%(10/12) in nasopharyngeal carcinoma tissues and23.1%(3/13) in control groups,NNMT protein positive stainning rate was41.6%(5/12) in nasopharyngeal carcinomatissues, and84.6%in control group (11/13); ANXA3protein expression rate washigher in nasopharyngeal carcinoma, while the positive rate of NNMT proteinexpression was lower in the control group, the differences were statisticallysignificant (P <0.05).CONCLUSION:1. The expression of ANXA3is increasing in nasopharyngeal NP69cells of lowexpression STGC3, whereas NNMT expression is decreased.2. The expression of ANXA3is increasing in nasopharyngeal carcinomal cellsCNE1, CNE2and nasopharyngeal carcinomal tissues, whereas NNMT expression isdecreased.
