Senile disease (ie, Alzheimer’s disease, AD) is one kind of chronicity degenerative nerve system diseases which has undetermined cause and feature by intelligence breakdown.In order to provide the theoretical and experimental evidences for effective prescription which is suitable to treat AD in TCM clinical practice and clarify the anti inflammatory reaction mechanism of AD, and also discussed the relative researches of treating AD mainly by invigorating the kidney from various theoretical and experimental levels.In the theoretical study aspect,according to the theory of kidney storing essence, the brain being the reservoir of the marrow,using the liver and kidney theory of TCM and the relationship between kidney and brain,the paper probed thoroughly into what important effects kidney deficiency coagulated phlegm blood stasis have in the mobility process of AD. We established the basic constitution of treatment to AD,which was developed the Modified Sanjiasan decoction to treat AD. At present,a number of studies have proved that AD is one kind of the central nervous system inflammatory reaction,basing on the past effective results of clinical and animal studies,we designed the cell experiment to take further step to reveal the possible anti inflammatory reaction mechanism how the method of the treatment acted.In the experimental study aspect, we developed cultured hippocampal neurons injury model induced by β-anyloid25-35protein(A β25-35) dealed with aging. Primary cultured hippocampal neurons were divided into below groups:normal group,model group,huperzine a group,low dose of TCM group,moderate dose of TCM group and high dose of TCM group. We could observe the treatment effect of modified Sanjiasan decoction on the disease indexed by several inflammatory factors.It could be measured the content of IL-1α、IL-1β、 IL-6、TNF-α、TNF-β and NF-κB in the culture supernatant of cells by using the metheods of enzyme-linked immunosorbent assay(ELISA).The results showed that modified Sanjiasan decoction could reduce the content of IL-1α、IL-1β、IL-6、TNF-α、TNF-β and NF-κB in the culture supernatant of cells. It indicated that modified Sanjiasan decoction could have some apparent anti-inflammatory effect in cultured hippocampal neurons injury induced by A β25-35.The possible effective mechanism was that TCM inhibit the inflammatory response caused by Aβ disposition and reduce the nerve cells’ susceptibility to Aβ to reduce the release of cell inflammatory factors IL-1α、IL-1β、IL-6、TNF-α、TNF-β and NF-κB so as to decrease the neurotoxic effect in nerve cells to protect hippocampal neurons.
