Objective:1. To observe the expression phosphorylated Tau protein and P70S6K levels ofnormal control group,Alzheimer’s disease group,type2diabetes group,type2diabeteswith Alzheimer’s disease group in rats’ inhippocampus.2. To explore the role of P70S6K in type2diabetes mellitus and promote themechanism of the risk of Alzheimer’s disease.Methods:Male SD rats50,weight180to220g,were randomly divided into normal controlgroup(11rats),Alzheimer’s disease model group(AD group12rats),type2diabetesmodel group(T2DM group12rats) and type2diabetes with Alzheimer’s diseasemodel group(T2DM+AD group15rats).Keeping T2DM and T2DM+AD group ratshigh suger high fat diet,and1%streptozotocin(30mg/kg)intraperitoneal injectionaccording to body weight,the control group was given injection dose of citrate buffersolution.The blood glucose(FBG)≥16.7mmol/L as standard.To the hippocampus ofAD and T2DM+AD group rats injected with condensed Aβ1-40was established bystereotaxic apparatus,and Morris water maze test determine the success of modelingAD.Rats were breaking neck to death,gotton their hippocampus and blood.DetectingFBG,TG,TC,and observing the differnce of learning and memory ability of rats in4groups.Expression levels of phosphorylation of Tau protein and P70S6K were alsodetected by immunohistochemistry;usingRT-PCR detection total Tau protein mRNAand P70S6K mRNA.Results:1. The leverl of FBG,TG,TC in T2DM and T2DM+AD group rats were higherthan normal control group and AD group rats (P<0.01).AD and T2DM+AD grouprats’ learning and memory ability were worse than that of the normal control groupand T2DM group (P<0.01),there was different between T2DM+AD group and ADgroup (P<0.05).2. Immunohistochemistry: the expression of of phosphorylated Tau protein in AD and T2DM+AD group rats’ hippocampus increased more significantly than thatin normal control group and T2DM group rats (P<0.01),there was different betweenT2DM+AD and AD group (P<0.01).The expression of P70S6K in hippocampus ofT2DM+AD and T2DM group rats were more than the normal control group and ADgroup (P<0.01),there was significant difference between T2DM+AD and AD group(P<0.01).3. RT-PCR:The Tau protein mRNA expression of AD ang T2DM+AD grouprats’ hippocampus was more significantly enhanced than that of the normal controlgroup and T2DM group(P<0.01), there was different between T2DM+AD and ADgroup (P<0.01).Compared with normal control group,AD and T2DM group rats, theexpression of P70S6KmRNA in hippocampus of T2DM+AD rats enhanced(P<0.05);there was significant difference between the AD and T2DM group (P<0.01).Conclusion:In the4model groups, the expression level of phosphorylation of Tau proteinand P70S6K in type2diabetes mellitus with Alzheimer’s disease model group rats’was higher, The P70S6K may take a part in the mechanism that is type2diabetesincreased the risk of developing Alzheimer’s disease.
