| Objective:To obtain the dose-effect equation between the different concentrationsof nitrite solution and methemoglobin formation rate, and to invent a rapiddetection card for blood methemoglobin concentration in human by usingthe RGB color model.Methods:To acquire the dose-effect equation between the differentconcentrations of nitrite solution and methemoglobin formation rate, bloodsamples from healthy donors were incubated with different concentrationsof nitrite solution. According to the dose-effect equation, differentconcentrations of nitrite solution were obtained for generatingmethemoglobin blood samples of different percentage concentrations (10%to80%). Then, blood samples from healthy donors and methemoglobinblood samples of different percentage concentrations were dropped on theblood collection card. The values of red, green, and blue (RGB) code andHEX Code were acquired by RGB Color Finder software, and different methemoglobin color-pieces were generated. Finally, the rapid detectioncard for blood methemoglobin concentration in human was invented.ResultsThe relation between the different concentrations of nitrite solution (X)and methemoglobin formation rate (Y) was linear correlation and thedose-effect equation was: Y=2.12598+0.7123X (R2=0.9982,P<0.01). Togenerate methemoglobin blood samples of different percentageconcentrations (10%to80%), the concentrations of nitrite solution were11mmol/L,25mmol/L39mmol/L,53mmol/L,67mmol/L,81mmol/L,95mmol/L,109mmol/L.According to the methemoglobin blood samples of differentpercentage concentrations (10%to80%), the RGB values for differentpercentage concentrations were:(205.10.16),(193.33.35),(183.34.38),(170.35.39),(157.36.41),(144.37.43),(133.38.44),(119.39.48),(103.40.49)and the HEX Code values are:(#CD0A10),(#C12123),(#B7226),(#AA2327),(#9D2429),(#90252B),(#85262C),(#772730),(#672831).Compared with blood samples from healthy donors, the colors of the bloodcollection card were gradually darkened until turing to brown as thepercentage concentrations of methemoglobin increased.ConclusionThe rapid detection card for blood methemoglobin concentration inhuman probably might be used for forensic routine cases, criminal investigation and clinical doctors. This detection card is accurate, fast,secure and cheap, therefore, it should be popularized in the future. |
