Object To investigate the effects and possible mechanisms amongBMP9expression, migration and invasion of lung cancer A549cells invitroMethods Immunohistochemistry staining(IHC) was used to detectthe expression of BMP9and IL6in clinical lung adenocarcinoma andadjacent tissues of lung adenocarcinoma, Human lung cancer A549cellswas used and the over-expression of BMP9was infected by adenovirusBMP9in vitro, RT-PCR and Western blot were used to detect theexpression of BMP9in recombinant AdBMP9/A549cells; to detect thechanges of the recombinant AdBMP9/A549cells invasion and migration,cell wounding assay, Transwell invasion assay were carried out. At last,RT-PCR and Western blot were used to detect the expression of IL6, Theexpression of total Akt and phosphorylation Akt protein were detected bywestern-blot after BMP9adenovirus infection.Results the expression of BMP9and IL6protein was higher in lungadenocarcinoma tissues than adjacent normal tissue, it has statisticalsignificance(p<0.05)。BMP9mRNA and protein were expressed inrecombinant AdBMP9/A549cells; the healing rate of experimental groupwas increased from (85.4±2.1)%and (86.5±3.4)%in control group to (97.4±2.6)%(P <0.05), the number of invasion cells which moved acrossthe matrix barrier in experimental group was increased from (105.5±13.1)and (113.3±14.9) in control group to (224.3±24.6)(P<0.05). In comparisonwith the control groups, the expression of IL6and phosphorylation Aktwere significantly increased in AdBMP9/A549group. Conclusion BMP9can promote the invasion and migration of lungcancer A549cells possibly through up-regulation of IL6expression andactivate of the PI3k/Akt cell signaling pathway.
