Background:Lung cancer has become one of the highest malignant diseases in morbidity andmortality. Our country is the most populous country in the world with lung cancerpatients, with a growth of4000thousand new lung cancer cases one year. Thetraditional treatments for lung cancer are composed of surgery, chemotherapy andradiation therapy, which have been up to platform period.5-year survival rate castmaintains in a low level. As the molecular mechanism of research for lung cancer, theepidermal growth factor receptor-tyrosine kinase inhibitor(EGFR-TKI) opened aprelude of molecular target therapy.The treatment target of EGFR-TKI is the epidermal growth factor receptor(EGFR)protein. EGFR has tyrosine kinase activity and integrates the epidermal growth factorinto phosphorylated epidermal growth factor receptor(pEGFR). They can activate aseries of signaling pathways in the downstream, one of which is the way forphosphotylinosital3kinase (PI3K)and serine threonine protein kinase,(AKT). Thepathway activates the intranuclear related gene and lead to tumor cells propagation、apoptosis suppression、the proliferation of cells factor synthesis、tumor cells totransfer and concurrent chemoradiation resistance.The method ofimmunohistochemical(IHC) was used to measure the protein expression of EGFR,pEGFR、PI3K and AKT in the cases of non-small cell lung cancer(NSCLC), and therelationship between the clinical and pathological features were analized to explor theconnection between EGFR and the pathway of PI3K/AKT and to understand therelationship between occurrence,development and treantment of NSCLC..Purpose:To research from the protein levels of EGFR、pEGFR、PI3K and AKT the expressionsof protein in lung cancer and normal paracancerous tissues, and to analyze the relationship of lung cancer pathological points type、the clinical stages、lymph nodemetastasis and tumor differentiation degree.To find the role of EGFR、pEGFR、PI3Kand AKT in NSCLC.Method:100cases of surgical operation of lung cancer and their corresponding normal tissueadjacent were fixed with formalin liquid, paraffin-embedded, cut into4μm thick serialsections. The types of all the specimens were confirmed by pathologists. The methodof IHC was used to detect the expression of EGFR, pEGFR,PI3K and AKT protein inNSCLC and normal paracancerous tissues. SPSS software was used to perform achi-square test positive and P <0.05was a statistical significance.Results:In100cases of NSCLC, positive expressions rate of EGFR, pEGFR, PI3K and AKTprotein were63%(63/100)、30%(30/100)、51%(51/100)、23%(23/100) respectively,and were significantly higher than that in100cases of adjacent normal lung tissuewith much lower positive expression rate of13%(13/100)、2%(2/100)、11%(11/100)、7%(7/100) seperatetly (each P<0.05). there was no significant correlation in theexpression of EGFR, PI3K and AKT with different age, sex, smoking index,differentiation degree and the lymph node metastasis(P>0.05), but the differencebetween the tissue types was significant(P<0.05). The expression of pEGFR wasrelated with sex, smoking and tissue types(P<0.05) but was not related with age,differentiation degree and the lymph node metastasis(P>0.05).Conclusion:The positive expression rate of EGFR, pEGFR, PI3K, and AKT protein in100casesof NSCLC tissues was significantly higher than the corresponding tissue adjacent tocancer, and EGFR, pEGFR, PI3K, and AKT protein were related with the occurrenceand development of lung cancer. EGFR had no relationship with pEGFR. EGFR hadno relationship with PI3K/AKT pathway.
