Multiple myeloma is a kind of abnormal proliferate disease of plasma cells in bone marrow, which shows a characteristic of accumulation of malignant plasma cells in bone micro-environment and great amounts of immune globulin produced by abnormal expression in bodies. Clorf35gene is a MM malignant related genes, which is selected by JingYan Tian through clone expression from ARH-77Gene Pool. It locates in1q42.13, its full length of cDNA is1300bp, including8exons, and expression product is about29kD. After the gene was transfected into NIH3T3cells, it seemed that cell proliferation enhanced obviously and cell cycles tended to transfer from G1phage to S phage by flow cytometry. At the same time, C1orf35gene transfection to COS-7cells after laser confocal technique showed that the gene encoding protein located in the nucleus. Bio-information analysis displayed that C1orf35gene may be as a transcription factor and play the role of transcription regulation.ARH-77belongs to the plasma myeloma cell lines, this study adopted lentiviral vector GV248/C1orf35shRNA to infect ARH-77cells in order to build stable cell lines expressing ARH-77/C1orf35-knockdown and knocked down the expression of C1orf35gene in ARH-77cells. After C1orf35gene knocked down, the expression of c-myc gene as well as ARH-77cells proliferation was significantly decreased. This shows C1orf35gene may have some relation with c-myc gene, leading to synergy of pathogenic in multiple myeloma. In the western blotting experiments, compared with protein expression of Cyclin E and CDK2in ARH-77cells, which are closely related to the cycle of cells and promote cells to transfer from G1phage to S phage, ARH-77/C1orf35-knockdown is significantly decreased. This means C1orf35gene promoting cell proliferation may be through the regulation of cell cycle related protein. As for how C1orf35genes involve in cell regulation, further research about C1orf35genes involving in cell signal transduction is required.
