| Multiple myeloma is a kind of abnormal proliferate disease of plasma cells in bone marrow, which shows a characteristic of great accumulation of malignant plasma cells in bone micro-environment and amounts of immuneglobulin produced by abnormal expression functioning in bodies. The accumulation of immuneglobulin induced a myriad of clinical symptoms including bone destruction, anemia, immune deficiency, renal failure and hypercalcinema. Clorf35is a related gene got by selecting the Gene Pool of human plasma ARH-77cell line in a way of expression cloning. After transfect this gene into NIH3T3cell, it was appeared that the capacity of cell proliferation was accelerated and cell cycle transferred from G1phage to S phage by flow cytometry. However, after transfect this gene into COS-7cells, it indicated that the protein of Clorf35locates in cell nucleus analyzing by confocal laser scanning microscope and that Clorf35may be a kind of transcription factor through bio-information analysis and have a capacity to cause carcinoma revealed by a tumor-burdened experiment of nude mice.This research, using lentiviral vector to infect NIH3T3/Clorf35cell in order to make Clorf35silence in the cell, reveal the results through MTT and cell counting that NIH3T3/Clorf35cell’s proliferation is conspicuously descended after silencing the expression of Clorf35and the proliferation of MM cells is close related to this gene. Thus, there is of great significance deeply determining its definite mechanism to understand the pathogenesis of MM and discover targets for new drugs. |
