Andrographis paniculata Acanthaceae plant is one of the commonly used traditional Chinese medicine clinic, the bitter taste, slightly cold, lung, large intestine classics. With heat clearing and detoxifying, Liangxue swelling, a natural antibiotic reputation. Through the study found that the main effective component in Andrographis paniculata is two terpene lactone compounds takes andrographolide as representative, it has anti-inflammatory, hepatoprotective, anti tumor with swelling and pain etc.. Widely used in clinical pneumonia, influenza, dysentery and infectious diseases, the separation of active ingredients of Andrographis paniculata and andrographolide compounds obtained has certain anti-inflammatory activity as the premise, the extract of Andrographis paniculata, effective parts, andrographolide and its derivatives on rat asthma experiment, anti asthmatic effect of Andrographis and andrographolide derivatives.Using ovalbumin as allergen, with neutral Al (OH)3antigen prepared from gel to induce rat, inhaled1%ovalbumin solution, the preparation of rat asthma model. Given the Andrographis and andrographolide compounds were observed in lung tissue structure remain intact, airway wall thickening phenomenon is improved, surrounding the alveoli infiltration of inflammatory cells decreased significantly, mucosal edema, no mucus embolus.Leukocyte microscope count in rat lung lavage fluid (WBC), calculation of total number of eosinophils (EOS), neutrophil (N), macrophage (M), lymphocyte (L) percent (%), data show:the total number of rats in the model group (87.63±2.67) WBC and EOS%(11.76±1.03) was significantly higher than that in normal group, total WBC (8.88±1.25) and EOS%(1.30±0.28); andrographolide (A) the total number of WBC (60.75±2.60) EOS%,(6.25±0.24); total andrographolide derivatives DA WBC (58.25±3.11)EOS%,(6.02±0.17) lower than that of model group, but higher than that in the normal group, the difference was statistically significant (P<0.05); the effective parts of Andrographis paniculata (SEA) low dose group WBC (78.5±3.89), the total number of EOS%(9.48±0.35), medium dose group (71.87±3.36) WBC total, EOS%(8.39±0.27), the total number of WBC of high dose group (67.5±3.29), EOS%(7.66±0.43) compared with the model group was statistically significant (P<0.05); the andrographolide derivatives of-1(A-1) the total number of WBC (54.26±2.76) EOS%,(5.56±0.19) compared with the model group with significant difference (P<0.01); dexamethasone (Dex) group, WBC count (31.88±1.96) EOS%,(3.36±0.37) compared with the model group had significant difference (P<0.01).Lavage fluid of rat ELISA Kit (BALF) in inflammatory cytokines interleukin-1(IL-1), tumor necrosis factor alpha (TNF-α) and cytokine interleukin-2(IL-2), interferon gamma (IFN-γ) were detected, results show that the IL-1factor group (52.82±2.13) inflammation model and TNF-α (75.14±5.72) was significantly higher than that in normal IL-1group (12.94±2) and TNF-α (20.12±10.34), A group (30.79±2.82) IL-1and TNF-α(42.33±13.52); DA group of IL-1derivatives (29.86±4.05) and TNF-α (37.34±9.42) lower than that of model group, higher than that in the normal group, the difference was statistically significant (P<0.05); A-1group compared with the model group significantly decreased IL-1(27.63±3.03) and TNF-α (36.79±10.85) level, significant difference was statistically significant (P<0.01); Dex group IL-1(22.82±4.68) and TNF-α(32.32±9.76) significantly lower than those in the model group, the difference is statistically significant (P<0.01); immune factor IL-2lung lavage fluid of rats in the model group (234.41±26.54) of IFN-γ and (27.23±5.02) was significantly lower than that in normal IL-2group (496.01±83.53) and IFN-γ (74.1±10.02), A group (295.69IL-2±85.64) and (48.03±IFN-γ18.16); DA group (315.20±57.49) IL-2and IFN-γ (56.15±14.05) higher than that of model group, lower than the normal group, the difference was statistically significant (P<0.05); A-1group (324.55±87.89) IL-2and IFN-γ (57.64±28.61) compared with the model group had significant difference (P<0.01; SEA). Low dose of IL-2group (269.52±30.49) and IFN-γ (43.86±11.65); the middle dose group (302.83±42.44) IL-2and IFN-γ (45.88±32.84); high dose of IL-2group (371.71±66.82) and (57.55±30) IFN-γ differences compared with the model group was statistically significant (P<0.05) IL-2, Dex group (389.61±14.67) was significantly higher than that in the model group, the difference was statistically significant (P<0.01).This topic research Andrographis and andrographolide compounds anti asthmatic effects, the experimental data show that andrographolide anti asthmatic effect is good, and its derivative-1anti asthmatic effect is better than that of andrographolide, can significantly reduce the number of inflammatory cells and inflammatory factors, anti asthmatic effect. The effective parts of traditional Chinese medicine Andrographis paniculata by supramolecular enrichment technology has a certain role in the mitigation of asthma, with the increase of the dose gradient enhancement, anti asthmatic effect.
