Effects Of Danshen On The Airway Remodeling In Asthmatic Rat

Objective: To establish SD rat asthma model of airway remodeling and investigate the effect of Danshen on airway inflammation and remodeling. Concurrently, make a comparison with glucocorticoids, further to provide new experimental evidence for the prevention and treatment of airway remodeling in asthma.Methods: (1) Rat chronic asthmatic model of airway remodeling was established by extending OVA boosting time. Rats were sensitized and boosted by injection with ovalbumin (OVA) plus immune adjuvant (aluminum hydroxide gel) every eight days, challenged by an aerosol of OVA every two days. As a negative control, animals were exposed to saline instead of OVA inhalation. (2) 40 female SD rats of asthma were randomly divided into five groups: model group, early budesonide group (early BUD group, atomization of 5ml 0.02% budesonide), early Danshen group (early DS group, intragastric administration of 400 mg/kg.d), late BUD group (late BUD group, atomization of 5ml 0.02% budesonide), and late DS group (late TP group, intragastric administration of 400 mg/kg.d), each group had eight rats. Meanwhile, eight healthy female SD rats were as control group. Budesonide or Danshen were administered two weeks before molding in early BUD group and early DS group. By contrast, in late BUD group and late DS group, budesonide or Danshen were administered five weeks after molding. These following index were observed at the period of 12 weeks:â‘ cell population and classification of BALF (bronchoalveolar lavage fluid, BALF),â‘¡lung tissue pathology( area of total bronchus wall, bronchial smooth muscle, and collagen deposition). hematoxylin-eosin (HE) staining and Masson staining were adopted.Results:(1) Compared with control group, there was a significant increase of BALF cell population, eosinophils (Eos), neutrophils (PMN), and epitheliums in asthmatic model group, and early administration of Danshen or budesonide had remarkable downregulated effect on the cell population, but no difference was observed between early Danshen and budesonide groups. Concurrently, the cell population was also decreased in late Danshen and budesonide groups; however, the change characteristics was a little different from early intervention, the decrease of PMN in late Danshen group was more obvious than that in late budesonide group, the changes of BALF cell population, Eos and epitheliums had no significant difference between two groups. The effects of early Danshen or budesonide administration were superior to late administration. (2) Lung pathology showed that asthmatic rats manifested coarctation of airway lumen, desquamation of bronchial epitheliums, thickening of tunica mucosa bronchiorum, increase and extension of bronchial folds, thickening of airway smooth muscle and alveolar septum, increase of collagen deposition. (3) Distinct increase in area of total bronchus wall, bronchial smooth muscle, and collagen deposition was found in asthmatic models, compared to control group. Early intervention could reverse these effects to some extent, regardless of Danshen or budesonide; however, there was no significant difference between these two groups. In contrast, administration of Danshen in late stage got the advantage over budesonide in late stage, though both also had positive effects.Conclusions: (1) Sensitization and aerosol of OVA can successfully establish chronic murine asthmatic model. (2) Airway inflammation could be mitigate by using Danshen and budesonide, early intervention is superior to late intervention. (3) No difference in mitigation of airway inflammation was observed between early Danshen group and early budesonide group. However, in late intervention, the effect of Danshen on PMN infiltration is better than budesonide. (4) Airway remodeling of murine asthma could be effectively mitigated and postphoned by early intervention of Danshen or budesonide. However, administration of Danshen in advanced stage can get the advantage over budesonide.
