Objective: The aim of this study was to detect the expression of Notch1、vascularendothelial growth factor (VEGF)and CD31in the cutaneous squamous cellcarcinoma(CSCC) of head&face, and calculate the microvessel density(MVD).Thisexperiment investigated the effections of Notch pathway and VEGF pathway inCSCC and the relationship with MVD.Materials and Methods: The expressions of Notch1,VEGF and CD31proteinwas detected by SP immunohistochemical methods in40cases of CSCC tissue ofhead&face and20cases of normal skin tissues. Quaque group experimenental datawere demonstrated with x s.Student’st test, Pearson Chi-square (χ2) test andSpearman’s rank correlation analysis were used perfoming the correlation betweenNotch1expression and VEGF expression. A probability (p) value less than0.05wasthe significant denominator for statistics.Results: By immunohistochemistry, the positive expression ratio of Notch1protein(25.0%) in CSCC was evidently lower than that (80.0%) in normal skin tissues, therewas a significant difference between the two groups(p<0.05*). Expressions of Notch1protein in grade Ⅰ-Ⅱgroups and Ⅲgroups were40%and20%respectively, therewas obvious difference between both of the groups(.χ2=4.181,p<0.05*). VEGF andCD31proteins were abnormally increased in CSCC as compared with normal skintissues. Expressions of VEGF and CD31in CSCC Ⅰ-Ⅱ groups were alsosignificantly higher than those in CSCC Ⅲgroups. MVD in Ⅰ-Ⅱgroups and Ⅲgroups were38.46±4.79and48.35±5.01respectively, which remarkably higher than19.83±2.59in normal skin tissues. In40cases of CSCC tissues, MVD in caseswith Notch1-negative group was higher compared to the positive group(80.51±4.11vs40.59±5.43,t=-26.215, p<0.001*). There was obvious negative correlation betweenthe expressions of Notch1and VEGF proteins with the correlation analysis.Conclusion:1) Expression of Notch1protein in CSCC was lower than normal skin tissues,furthermore, to submit negative correlation with histological grade of CSCC andthe value of MVD in CSCC.2) VEGF protein expression in CSCC was significantly higher than nomal skin tissues,furthermore, and VEGF submit positive correlation with histological grade ofCSCC and the value of MVD in CSCC.3) There was obvious negative correlation between the expressions of Notch1andVEGF proteins.
