Determintion For The Content Of Puerapin Injections,in Vivo Metabolism And The Effects On CYP2E1Activity

This dissertation is composed of three chapters. The first chapter determinted the content of puerarin in three kinds of puerarin injections by HPLC method. The second chapter researched absorption and metabolism of puerarin in rats by HPLC. The third chapter analyzed effects of puerarin on rat hepatic microsomal enzymes CYP2E1by probe drugs method in vitro. The fouth chapter analyzed of the domestic market of Puerarin injection status.Chapter1. Determintion for the content of puerarin in puerarin injections by HPLC MethodA Merk, Purospher STAR RP-C18column was used to isolate at29℃and mobile phase was methanol and1%glacial acetic acid, water with the flow rate of1.0mL·min-1, detection wavelength was250nm. The puerarin was separated with a good linearity in the sample size range of0.192～1.152μg. The average recovery was99.61%, with RSD1.43%. The results showed that puerarin in three kinds of injection could be well separated under the selected conditions, had good linear relationship. The method was simple and quick, reasonable and could provide scientific basis for the quality control of puerarin injection.Chapter2. Research absorption and metabolism of puerarin in ratsThe plasma concentration of puerarin was determined by HPLC collected blood from tail vein. The pharmacokinetic parameters was calculated with PK Solver2.0program. To establish a RP-HPLC method for the determination of puerarin in rat plasma, and research absorption and metabolism of puerarin after rat tail vein injection in240minutes. The results were pharmacokinetics of puerarin could be fitted to two-compartment model in rats. It showed that the distribution and elimination of puerarin were faster, with low toxicity, quick metabolism and hard accumulation advantages. Chapter3. Analyze effects of puerarin on rat hepatic microsomal enzymes CYP2E1in vitroPuerarin was analyzed by HPLC useing rat liver microsome by ultracentrifugation, and incubate after suitable incubation conditions. The results showed that20min,30min,60min blank and experimental groups were not found4-nitro catechins. It showed that puerarin without induction and inhibition effects on rats stem cells CYP2E1.Chapter4.
