ObjectiveLung cancer has become the leading cause of cancer-related death, and theprevalence rate increased year by year. The early diagnosis is faced with manydifficulties. Drugs in the treatment of lung cancer is still the main means, while theside effects of traditional chemotherapy drugs is apparent, and the targeted drugs hastoo many individual differences. In recent years, research found that curcumin hasbetter antitumor, anti inflammation, anti-oxidation effect, at the same time, the sideeffect is small, but the bioavailability is poor, so the curcumin analogues has greatresearch value, becoming one of the breakthrough point of the development of newantitumor drugs.To explore the ability of curcumin analog Sunwsz08to induceautophagy in A549cells as well as inhibit their proliferation.MethodsA549cells were incubated with various concentrations of Sunwsz08for differentdurationsand their morphology was examined using an inverted microscope. TheA549cells were stained with acridine orange and propidiumiodide,and theirmorphology was further examined by fluorescence microscopy.In order to examinethe ability of Sunwsz08to promoteautophagy inA549cells,GFPLC3positioning was used to identify autophagic vacuoles.The MTT assay was used to assess thecompound′s ability to inhibit cellular proliferation.ResultsAt concentrations of8and10μmol/L,Sunwsz08significantly increasedautophagy in A549cells.At concentrations above16μmol/L,the analog caused anincrease in the number of cell vacuoles，and it induced abnormal cell morphology andcell death.Sunwsz08also inhibited A549proliferation in a dose-and time-dependentmanner.Conclusion1.Curcumin analogues of Sunwsz08can inhibit the proliferation of A549cells, and theeffects increased with prolonging time and concentration, The IC50of12h and24h were35.37μ mol and32.35μ mol/L2.Sunwsz08can directly induce A549cell death.3. Sunwsz08can induce autophagy in A549cells.
