ObjectiveHeart failure remains the leading cause of death worldwide,causing a significantburden on health care systems across the globe,especially in the aged.4.1R can affectthe positioning of ion channels and regulating cell connections via cellular adhesivemolecular.Through this study,we want to discuss the expression of4.1R in myocardialcell of aged mice and association between4.1R and heart failure.Methods48old male mice were randomly divided into2groups.HF group(n=26):Regularfeeding and abdominal subcutaneous injecting of isopropyl adrenaline (ISO)5mg/kg/d,14days.Control group(n=22):Regular feeding and abdominal subcutaneousinjecting of normal saline (NS) for14days.After4weeks,all the mice were killed.Thebody weight and left ventricular weight were measured;left ventricular weight indexwere calculated;the pathological changes of the heart were observed;4.1R inmyocardial cell were analyzed by immunohistochemistry and immunofluorescence.The statistical data was analyzed by t-test,rank sum test and Pearson correlationanalysis.Value of P<0.05was considered to be significant.Results1.Comparison of left ventricular mass index(mg/g):In the control group,theindex was2.84±0.02;in the HF group,the index was4.71±0.02.Compared with the control group,the left ventricular mass index of the HF group increased obviously(P<0.01).2.Pathological changes:The myocardial tissue pathological score in the HFgroup increased obviously compared with the control group(P<0.01).3.Expression of4.1R in myocardial cell:4.1R locates in the plasma membrane ofthe cell.4.Change of4.1R:The positive rate of4.1R in the control group and HF groupwere6.35±0.72and11.31±0.98respectively.The positive rate increased in the HFgroup significantly (P<0.01).5.Linear correlation analysis:In the HF group,the positive correlation was foundbetween the left ventricular mass index and the positive rate of4.1R(r=0.745,P<0.01).Conclusion1.Isoprenaline can induce myocardial hypertrophy and heart failure.2.4.1R may locate in the plasma membrane of myocardial cell.3.4.1R may increase in the heart failure of old mice.It may guide the treatmentand prognosis of heart failure.
