BackgroundEsophageal carcinoma is one of the most common malignant tumors inChina,with high morbidity, mortality and serious damage.The esophageal carcinomacan be divided into squamous cell carcinoma and adenocarcinoma.In the west, themorbidity of esophageal adenocarcinoma increases distinctly, and has become themain pathological type of esophageal carcinoma in the world at present.In our country,the squamous cell carcinoma is the main pathological type of esophageal carcinomaand the morbidity increases daily, but adenocarcinoma with less morbiditycomparatively. The occurrence and development of esophageal carcinoma is acomplicated process with the interaction of multiple gene and multiple steps. Inrecent years, many oncogene and tumor suppressor genes with abnormal expressionare found in esophageal carcinoma.Recent researches suggest SASH1is a member of the SLY gene family whichconnects code with protein. FBXO32is classified in the FBX protein family, both ofthem are new candidate tumor suppressor genes, and take parts in the biologicalbehavior of many malignant tumors.The reducing expression of SASH1and FBXO32in colorectal cancer, gastric cancer, lung cancer, melanoma, glioma and other tumors relates to infiltration, transfer and prognosis of tumor. The relationship betweenSASH1,FBXO32and squamous cell carcinoma is rarely researched at home andabroad. To clear the relationship of SASH1and FBXO32protein and squamous cellcarcinoma will contribute to the development mechanism of squamous cell carcinoma,and maybe provide new test evidence for the diagnosis of squamous cell carcinomaand new target of gene therapy.ObjectiveBy testing the expression level of SASH1protein and FBXO32protein in humanesophageal squamous cell carcinoma and normal esophagus mucosa tissue, anddiscussing the clinical significance, provide theoretical basis for monitoring,development of prognosis and targeted therapy.MethodsWe chose76cases of esophageal carcinoma specimens and the correspondingnormal tissues adjacent to the tumor (more than5cm apart from the edge of tumor)performed in the Department of thoracic surgery operation of the First AffiliatedHospital of Zhengzhou University from February2012to march2013, tested theexpression level of SASH1protein and FBXO32protein by S-P method, analyzed theresults by χ2test and Spearman’s rank, the test level was α=0.05, and discussed therelationship between SASH1protein, FBXO32protein and clinical pathologicalfactors of the patient with esophageal squamous cell carcinoma.Results1.The positive rate of SASH1protein in esophageal squamous cell carcinoma was35.52%, and it was53.95%in normal tissues adjacent to the tumor. So the positiverate of SASH1protein was lower in esophageal squamous cell carcinoma than normaltissues adjacent to the tumor.The difference was statistically significant(χ2=4.694,P <0.05).The TNM stage and differentiation of SHSH1and esophageal squamouscell carcinoma in esophageal squamous cell carcinoma relate with the lymph nodemetastasis(P <0.05), nothing to do with gender and age(P>0.05).2.The positive rate of FBXO32protein in esophageal squamous cell carcinoma was36.84%,and it was55.26%in normal tissues adjacent to the tumor. So thepositive rate of FBXO32protein was lower in esophageal squamous cell carcinomathan normal tissues adjacent to the tumor. The difference was statistically significant(χ2=4.447,P <0.05).The TNM stage and differentiation of FBXO32and esophagealsquamous cell carcinoma in esophageal squamous cell carcinoma relate with thelymph node metastasis(P <0.05),nothing to do with gender and age(P>0.05).3.The expression level of SASH1protein in esophageal squamous cell carcinomawas positively correlated with that of FBXO32protein(r=0.300,P <0.05).Conclusions1.The low expression of SASH1and FBXO32in esophageal squamous cellcarcinoma may have some relationship with the infiltration, growth and distantmetastasis of esophageal carcinoma, hopefully to be an important indicator of thediagnosis of esophageal carcinoma, new target of gene therapy and diagnosis ofprognosis.2.The expression level of SASH1protein in esophageal squamous cell carcinomais positively correlated with that of FBXO32protein, some synergistic action mayexist in the occurrence and development of esophageal carcinoma.
