Background and ObjectiveSquamous cell carcinoma and Basal cell carcinoma are common malignanttumors of skin tissue. They have the same origin, predilection site and pathogenesisof similar, and two kinds of diseases in the pathogenesis of local site also have similarclinical manifestations. But the transfer occurs in tumors of two kinds of disease hassignificant difference.Proteomics refers to the proteome of cells or tissues are studied as awhole,analyzing the composition of the protein in the group composition and theeffects of interaction between them. The research of proteomics is mainly in the threeaspects.They are the proteins composition in the proteome, differential proteomicsstudy and proteomics and the role between the various proteins in the biologicalactivities.This study use relevant technology of proteomics to analysis the extrictedproteins in the human skin squamous cell carcinoma and basal cell carcinoma cell,and to find out the differences of protein, and the functional verification of someproteins. It will have signifficance to the cutaneous squamous cell carcinoma andbasal cell carcinoma, in early diagnosis and early metastasis, tumor marker identification, treatment of tumor specific sites of screening, and the development ofclinical new treatments.Methods1Cell culture and extraction of sample protein,In this experiment, the humanskin squamous cell carcinoma A-431cell line and human basal cell carcinomaTE-354.T cells cultured for routine for tumor cell model. Two kinds of cells inlogarithmic growth phase, the cells adherent to the culture flask cultivation area of the90%samples, protein extraction.And the Bradford method is applied to the sampleprotein concentration was measured.2Two-dimensional polyacrylamide gel electrophoresis sample protein,Through the IPG dry strip of second time hydration, isoelectricfocusingelectrophoresis, balanced IPG glue, vertical SDS-PAGE electrophoresis,colorand other steps are protein separation on the samples of two kinds ofproteins, eachsample are respectively3pieces of two-dimensional electrophoresis plywood, andapplication of image scanner to scan, two-dimensional gel electrophoresis of proteinsamples obtained from two. Software for analysis of two-dimensional gelelectrophoresis image analysis,find out the differential protein spots.Results1Two-dimensional electrophoresis map of the two cell lines extracted proteinstube are basically same, each protein as basic gathered at the isoelectric point of pH4~7range; molecular weight were in the range of15kDa~150kDa.2Through the analysis of two-dimensional gel of samples extracted protein inskin squamous cell carcinoma cell line and skin basal cell carcinoma cell line in, thereis a difference of16.8%protein spots, including13protein in squamous cellcarcinoma cells were up-regulated and2down regulated protein expression insquamous carcinoma cells.4protein spots in basal cell carcinoma group was notdetected. Conclusion1There are some differences between two-dimensional gel electrophoresis ofsamples extracted protein in skin squamous cell carcinoma cell line and skin basalcell carcinoma cell lines. Skin squamous cell carcinoma containing protein fractionsare mor complex than the basal cell carcinoma.2The differentially expressed proteins may be the cause of the influential factorson different aspects of metastatic squamous cell carcinoma and basal cell carcinomain tumor. Some of these proteins’ molecular weight and isoelectric point are(22.32,3.95)\(27.48,4.3)\(58.96,7.45)\(64.8,5)\(121.8,5.84)\(135.13,6.44).
