Objectives: This study used Qingfei jiedu decoction to intervene the model ofacute lung injury (ALI) in mice induced by lipopolysaccharide,to determine thecontents of the mice inflammatory factors PCT and TNF-αof blood serum,and toobserve the morphological changes of lungs in mice, to evaluate theanti-inflammatory effects of the Qingfei jiedu decoction on acute lung injury model inmice, to provide experimental basis for clinical medicine.Methods:60healthy BALB/c mice were divided into5groups randomly: thenormal group, the model group, the Qingfei jiedu decoction low-dose group, theQingfei jiedu decoction high-dose group, the dexamethasone group,12mice in eachgroup. except the normal group, all the other4groups were given intranasal with LPSto establish the ALI model.After2hours modeling the mice of normal group andmodel group were given normal saline l0ml/kg by gavage once per day;Qingfeijiedudecoction32.5g/kg,19.5g/kg were administered respectively to high-dose group andlow-dose group by gavage once a day;the dexamethasone group were givendexamethasone1mg/kg by gavage once a day. The five groups were all administeredfor3days. Before modeling and during treatment,measure the ear temperature in miceat9am. After the last drug to kill the mice and get the blood.Observe theappearance of their lungs and Weigh the body weight and lung weight,calculate thelung index.After HE staining, we observed the morphological changes of lungs withlight microscopy,and to score.To determine the levels of the serum PCT and TNF-α byELlSA.Results:(1) Observation of the general condition:there were2deaths during theexperiment; the mice of normal control group were lively and active,their furs were bright,and they have eating normally;the model group appears reducedactivity,Curly furs and no gloss, poor mental,lack of appetite;the Qingfei jiedudecoction high-dose group and low-dose group is better than the model group atactivity, the glossiness of furs, mental state and diet.The general condition ofdexamethasone group were similar to the Qingfei jiedu decoction groups.(2)Comparison of serum PCT and TNF-α: compared with normal group,the modelgroup’s concentration of serum PCT was higher (P<0.01);compared with model group,the concentration of serum PCT in the Qingfei jiedu decoction low-dose group, theQingfei jiedu decoction high-dose group and the dexamethasone group was lower (P<0.05).The concentration of serum TNF-α in model group was higher than that innormal group(P<0.01);compared with model group,the concentration of serum TNF-αin the Qingfei jiedu decoction low-dose group, the Qingfei jiedu decoction high-dosegroup and the dexamethasone group was lower(P <0.05).And both the concentration ofserum PCT and TNF-α were no significant difference between the Qingfei jiedudecoction low-dose group and high-dose group(P>0.05).(3) Comparison of lungindex:compared with normal group,the other4groups’ lung index rate was higher(P<0.01); compared with model group,the lung index of the Qingfei jiedu decoctionlow-dose group, the Qingfei jiedu decoction high-dose group and the dexamethasonegroup was lower(P<0.01).however,there was no significant difference between theQingfei jiedu decoction low-dose group, the Qingfei jiedu decoction high-dose groupand dexamethasone group.the difference between the Qingfei jiedu decoctionlow-dose group and high-dose group had no statistics significance(P>0.05).(4)Results of pulmonary tissue pathology and pathological integral:the normal groupmice’lung tissue structure were clear, alveolar were intact, alveolar interval wereuniform,and have no inflammatory cells.There is visible diffuse thickening ofalveolar interval and a large number of inflammatory cells(Necrotic neutrophil andmononuclear lymphocyte cells)in the model group; there is part of the alveolarinterval thickening and a small amount of inflammatory cells(mononuclearlymphocyte cells) in the Qingfei jiedu decoction low-dose group, the Qingfei jiedudecoction high-dose group and dexamethasone group.The pathological integral washigher in model group than that in normal group(P<0.01);compared with modelgroup,the pathological integral in the Qingfei jiedu decoction low-dose group, theQingfei jiedu decoction high-dose group and dexamethasone group was lower(P<0.01),however,there was no significant difference between the Qingfei jiedu decoction low-dose group,high-dose group and the dexamethasone group (P>0.05).Conclusion:(1) The Qingfei jiedu decoction can reduce the concentration ofserum PCT;(2)The Qingfei jiedu decoction can reduce the concentration of serumTNF-α;(3) The Qingfei jiedu decoction can alleviate the change of pulmonary tissuepathology.
