Endometrial carcinoma is one of the most common gynecologicmalignant tumor, it is a group of epithelium in endometrial malignant tumors,in its different origin of the tumor, the endometrial gland carcinoma is themost common. In our country, although the endometrial cancer incidence andmortality rate had no statistical data more accurate description, but at least wecan through the report, to understand each year the incidence of disease isincreasing, and the mortality rate is high. So, it is necessary to clear some andendometrial cancer occurrence, development related issues, to understandsome associated with tumor histological changes, and according to theabnormal proliferation of tumor cells, an important mechanism and theinvasion and metastasis of the surrounding tissue, tumor prognosis and riskassessment to determine more accurate, and rational treatment, so as to toenhance the treatment effect, improve the life quality of patients.Minichromosome maintenance proteins (Miniehromosome MaintenanceProteins, McMs) are a group of proteins, it is closely related with the DNAreplication. In twentieth Century80’s Bik-kwoon tye, found in the mutantyeast, and then use the experiment method of molecular and biochemicalextract, is an important group of homologous protein family, and they areclosely related to. Minichromosome maintenance protein (McMs) in thereplication licensing factor (replieationlieensingfactor, RLF) plays animportant role, is an important member, it combined with the chromatin in theinitiation of DNA replication, marking a new replication process started. Thiscompound as a "pass" DNA replication, has irreplaceable importance in thereplication of eukaryotic cells in DNA process. In the replication of eukaryotic DNA process, the MCM protein is regulated protein important to this process,and the DNA replication has resulted in the abnormal regulation of cellproliferation. High level expression of MCM protein in cell proliferation, celldifferentiation in quiescent or better in little or no expression. Thus the MCMprotein may be a specific marker for proliferating cells. MCM5is one of thesix members of the MCM family of proteins, that is to say, to detect theexpression of MCM5can indirectly understand the expression level of MCMprotein. Therefore, MCM5protein in tumor research process, become avaluable marker. Ki67is the proven and widely used as a cell proliferationmarker, it and the mitosis process is closely related to. especially on theprocess of tumor cell, combined with clinical data, confirm and malignanttumor staging, lymph node metastasis and prognosis correlation.Objective:The endometrial carcinoma (after operation) were used forimmunohistochemical staining method, detected the expression of MCM5protein, compared with that of normal endometrial tissue, and combined withclinical data, analysis of MCM5protein in the occurrence and development ofendometrial carcinoma, which provides a new method for the assessment tojudge the malignant degree and prognosis of endometrial carcinoma.Methods:Select the operation excision, and pathologically confirmed endometrialcancer tissue specimens from70patients (follow-up, through1years ofpostoperative recurrence in patients with29cases, compared with the controlwithout recurrence of41cases). Tissues from70patients with cancer typeswere endometrioid adenocarcinoma, no tumor types and clear cell carcinoma,serous carcinoma and other special organization. All patients were without anyspecial treatment, such as radiotherapy, chemotherapy and traditional Chinesemedicine, hormone treatment, record the patient name, age, tumordifferentiation, pathological stage (according to the2009FIGO staging),clinical data without lymph node metastasis, immunohistochemistry results,selected the same period of atypical hyperplasia, normal endometrial tissue samples were obtained from15patients as control. Expression of MCM5protein was detected by immunohistochemical S-P method in specimens.Application of statistical software SPSS for windows14, by χ2test and Ranksum test level data analysis.Results:1endometrial carcinoma detected by immunohistochemical SP method,the expression of atypical hyperplasia endometrial tissue, normal endometrialtissue protein MCM5: endometrial cancer and atypical endometrialhyperplasia total expression there was no significant difference in the rate (P>0.05), but relatively strong positive expression rate, the difference between thetwo was statistically significant (P <0.05).2endometrial cancer, endometrial hyperplasia expression of MCM5wider than normal endometrium, and has a significant difference (P <0.05).Conclusion:1. MCM5protein is highly expressed in endometrial carcinoma, and thedegree of tumor differentiation, stage, whether there is lymph node metastasis,MCM5in atypical endometrial hyperplasia tissues was significantly higherthan in normal tissue.2. MCM5protein is highly expressed in endometrial carcinoma andprognosis associated with endometrial cancer.
