| Objective:Through establish the solitary raising and chronic unpredictable mild stressmodel of depression.Observation of the Wuyou Decoction in chronic unpredictablemild stress and hippocampus brain-derived neurotrophic factor (BDNF) and itsreceptor tyrosine kinase B (TrkB) the expression of protein.Explore its mechanismand provide the theory for clinical medication.Methods:1. Model preparation: Using the following eight kinds of stimulationmethods.Include Water and food deprivation,Tail clip,Day and night reversed,Shockfoot,Level of shock,Ice swimming,Thermal stimulation.To prepare the rat model ofdepression.Randomly assigned a stimulus stress for a daily in21days.2. Feed on medicine in different groups: According to the group of the wholeamount of group simple random sampling is divided into six groups: the normalcontrol group, the model group, the high dose of Wuyou Decoction group, the middleand low dose of Wuyou Decoction group and Fluoxetine group, each group of10only.normal control group diet freely.Wuyou Decoction of high, middle and low dosegroup of9.84g/kg per day,4.9g/kg per day,2.46g/kg per day to fill the stomachmedicine, and model group daily fill the stomach2ml physiological saline, Fluoxetinegroup with16.3mg/kg daily fill the stomach to medicine. In the experiment date22for medicine for3weeks, once a day.3. Index detection: Each group before modeling, modeling, and after gavage test the behavioral indicators. After the end of the experiment using theimmunohistochemical methods to observe the effect on hippocampal CA1BDNF andTrkB expression of rat depression model. Using HE staining observe the number ofpositive cells and the changes morphology of cells in rat hippocampal CA1.Results:1. All the rats observed during normal adaptive feeding week, behavioraldetection without statistical significance(P>0.05).2. After21days of modeling, model group were generally larger changes, drycoat, reduced activity, unresponsive, horizontal scores and vertical scores decreasedthrough the open-field experimental significantly compared with the normal group (P<0.05), which show the depression model was successful.3. After21days of drug intervention the rat model of successful. Compared withmodel group, the horizontal and vertical scores in the Wuyou decoction high-dosetreatments,medium-dose treatments and fluoxetine treatments group was significantlyincreased(P<0.05).4. Compared with the normal group, the positive expression in model group ofBDNF and TrkB decreased (P<0.05);compared with the model group, Wuyoudecoction high dose group, middle dose group and fluoxetine group the expression ofpositive increased in hippocampal BDNF and TrkB (P<0.05).5. Compared with the normal group, the neuronal cell in CA1area ofhippocampus in model group was reduced and thin out, less cell nucleus have alreadydeformation and pycnosis via the HE stain.Conclusions:1. This paper uses the solitary raising and chronic unpredictable mild stressestablish rat model of depression, lay the foundation for the further experimentalresearch of Wuyou decoction.2. Wuyou decoction can improve the behavior characteristics of depression inrats with the most significant of high dose group.3. Wuyou decoction had antidepressant activity effect on chronic stress-induced depression model rats.The potential antidepressant effect may be related to increasethe level of BDNF andTrkB of hippocampal. |
