Objective:To observe the effects of Yunnan Dengzhanhua capsules on the renalfunction,the expressions of pro-inflammatory cytokines as interleukin-6(IL-6)、tumornecrosis factor-ɑ (TNF-ɑ),and fibrogenic factors as transforming growthfactor-β1(TGF-β1)、 plasminogen activator inhibitor-1(PAI-1) in the5/6nephrectomized chronic renal failure(CRF) rats,and to study the molecularmechanism of Yunnan Dengzhanhua capsules.Methods:①Modeling and Grouping:60SD rats were randomly divided intomodeling group(45rats)and normal control group(group A,15rats).Modeling grouprats were induced by5/6nephrectomized.The successful rat models were randomlydivided into model control group(group B),Benazepril does and the Dengzhanhuadoes groups(group C and D).There were13rats in each group.②Drug Administration:Normal saline was given to rats in group A and B bygastrogavage.benazepril(0.29mg/100g)was given to rats in group C bygastrogavage,while Dengzhanhua capsules(0.3g/100g)was given to rats in group Dby gastrogavage for12weeks.③I ndexes Testing:Scr、 BUN、 UPro/24h were detected before and aftertreating.Hematoxylin and eosin staining of pathological sections was used to observethe morphological changes.Expressions of IL-6and TNF-ɑwere determined byenzyme-linked immunosorbent assay analysis.Expressions of TGF-β1and PAI-1weredetermined by semi-quantitative analysis.Results:①Biochemical Indexes:Scr、BUN、UPro/24h of group B was significantly higherthan group A(P<0.05).Scr、BUN、UPro/24h of group C and D all decreasedsignificantly when compared with group B(P<0.05);While Scr、BUN、UPro/24h ofgroup D was lower than group C(P<0.05). ②:Real Time PCR Results:TGF-β1:Compared with group A,TGF-β1mRNA expression of group B washigher(P<0.05).TGF-β1mRNA expression of group C and D was significantly lowerthan group B(P<0.05),while TGF-β1mRNA expression of group D was lower thangroup C(P<0.05).PAI-1:Compared with group A,PAI-1mRNA expression of group Bwas higher(P<0.05).PAI-1mRNA expression of group C and D was significantlylower than group B(P<0.05),while PAI-1mRNA expression of group D was lowerthan group C(P<0.05).Compared with the benazepril,Yunnan Dengzhanhua capsulescould inhibiting the expressions of TGF-β1mRNA and PAI-1mRNA.③ELISA Result:IL-6:Compared with group A,IL-6expression of group B washigher(P<0.05).IL-6expression of group C and D was significantly lower than groupB(P<0.05),while IL-6expression of group D was lower than groupC(P<0.05).TNF-ɑ:Compared with group A,TNF-ɑ expression of group B washigher(P<0.05).TNF-ɑexpression of group C and D was significantly lower thangroup B(P<0.05),while TNF-ɑ expression of group D was lower than groupC(P<0.05).Enzyme-linked immunosorbent assay analysis showed significantdifference when compared with the benazepril, Yunnan Dengzhanhua capsules couldinhibiting the expressions of IL-6and TNF-ɑ.Conclusion:Yunnan Dengzhanhua capsules could reduce Scr、BUN and UPro/24h ofCRF rats induced by5/6nephrectomized,protect kidney function.It also couldpostpone the development of inflammatory status in CRF rats possibly throughinhibiting the expressions of IL-6and TNF-ɑ,and postpone the development of renalfibrosis possibly through inhibiting the expressions of TGF-β1and PAI-1,then slowdown the progress of chronic renal failure.This may be part of its effectivemechanism.
