The Research Of Mechanism On Regulated Th17Cells Of Wenyangtongluo Recipe On Collagen-induced Arthritis In DBA/1Mice

Objective: Discussion the effect of Wen Yang Tong Luo recipe （Juan biparticles） in vivo administration on the collagen-specific proliferative responses oflymphocytes and produced Th17cytokines (IL-17); reveal its mechanism on regulatedTh17cells of Wen Yang Tong Luo recipe on collagen-induced arthritis in DBA/1Mice,to provide theoretical basis for Wen Yang Tong Luo recipe cure RA.Methods: Established bovine type II collagen-induced arthritis (collageninduced arthritis, CIA) model in DBA/1mice, according to the clinical score wasaverage divided into pathological model group (normal saline), positive drug MTX(1mg/kg) group and Juan bi particle group (extract1.89g/kg),each group had10mice.The above groups give the medicine in9days after the attack,once a day, aboutlast20days. To observed the severity of arthritis on CIA mice with the method ofarthritis clinical scores and the pathological injury degree of arthritis in CIA mice withParaffin sections and H&E staining;Detecting proliferative responses of spleniclymphocyte in CIA mice with3H-TdR incorporation method to understand the stateof their cellular immune function;Detecting the expression levels of collagen-inducedsplenic lymphocytes Th17cytokines (IL-17A) by ELISA method.Results: Compared with the arthritis pathologic model group, Wen Yang TongLuo recipe（Juan bi particles）group were significantly reduce the clinical scores ofcollagen-induced arthritis in DBA/1mice, with a significant difference (P<0.05and P<0.01); HE staining results showed that,compared with arthritis pathologic modelgroup, the group Wen Yang Tong Luo recipe（Juan bi particles）could reduce the infiltration of inflammatory cells, prevented the damage of joint synovitis and joint; atthe same time, inhibited the collagen-specific immune response in CIA mice (P <0.01),reduced the produce levels of IL-17A in collagen-induced spleen lymphocytes (P<0.05).Conclusion: Wen Yang Tong Luo recipe has certain therapeutic effect oncollagen induced arthritis,The mechanism may be related to the suppression ofimmune, and down regulate the expression of IL-17A to play an anti-inflammatoryeffects.
