ObjectiveTo explore the influence of Jiao Tai Pill on P-glycoprotein and neurotransmitter Orexin A in rat sleep deprivation model.Methods50rats were randomly divided into five groups:control group, model group, Jiao Tai pi11group (5.5g/kg×d), berberine group (5g/kg×d), the West Pan group (0.04g/kg×d). We used the small platform water environmental method to establish rat model of sleep deprivation. Then we used real-time quantitative polymerase chain reaction (qRT-PCR) and Western blot to detect blood-brain barrier P-glycoprotein mRNA and protein expression. And enzyme-linked immunosorbent assay were used to detect brain parenchyma Orexin A expression.ResultsJiao Tai pill group’s P-glycoprotein mRNA and protein levels were significantly lower than the control group, model group and berberine group (P<0.05) Orexin A expression was also significantly lower than the other groups (P<0.05).GonclusionCombinatiom of cinnamon and berberine can enhance hypnotic effect, which is related to wake-promoting neuropeptide Orexin A expression. We believed that it could suppress the blood-brain barrier P-glycoprotein expression and promote berberine through the blood-brain barrier.
