Object i veQiongyugao is used to intervention the model of sub-acute aging rats caused by the function of D-galactose so as to analyzes Qiongyugao’s antioxidant effect on the rats model. Analyzed the network, pathway and gene ontology among different proteins in hypothalamus under the interv ention of Qiongyugao by a efficient and effective way by using bioinform atics techniques. Therefore we can ascertain the target protein of Qiong yugao acting on the hypothalamus to explain the mechanism of Qiongyugao at molecular level. We hope this study can lay a theoretical and experim ental basis for the later research.Methods423-month-old SD rats were randomly divided into3groups,14rats in each group. There are7male and7female rats in each group that wer e housed separately, marked as blank group(male), blank group(female), m odel group(male), model group(female), Qiongyugao group(male), Qiongyuga o (female). From the first day, the model group and the QG group were gi ven D-galactose (1.25mg·kg-1·d-1) intraperitoneal injection, and blank group were given isodose normal saline. At the same time, Qiongyugao gro up received Qiongyugao(214g·kg-1·d-1) by gavage, the blank group and mod el group were given isodose distilled water by gavage too. Those medicin e were consecutive used for8weeks. Got serum and liver after stopping drug to measure the total antioxidant capacity, glutathione peroxidase a nd catalase activity by ultraviolet spectrophotometry. Analysis differential protein annotation us ing clustering for Gene Ontology, analysis effect of differentially expr essed proteins network by using Ingenuity Pathway Analysis software, ana lysis the major metabolic pathway of differentially expressed proteins b y KEGG database.Results1.Get the model of subacute aging SD rats.2. Qiongyugao can improve CAT activity in male experiment aging rats’li ver, and had no significant difference in female rats’liver. The activ ity of GSH-PX in liver in male and female rats were increased, but no st atistical significance. The activity of serum T-AOC of male and female a nd male rats were significantly higher than model group, has statistical significance.3. Among the GO comments of differentially expressed proteins,68associa ted with biological process,33were related to cell composition, and29have a bearing on molecular features.4.There are2or more nodes may be the important protein in differential ly expressed proteins of Qiongyugao in the network map. They are ubiquiti n C; phosphatase and act in regulator; ubiquinol-cytochrome C reductase c ore protein1; protein phosphatase1regulatory (inhibitor) subunit13like; internexin neuronal intermediate filament protein,alpha; NADH dehy drogenase (ubiquinone) Fe-s protein1; N-ehtyimaleimide sensitive fusion protein attachment protein, beta; ARP3actin-related protein3; succinat e dehydrogenase complex, subunit A, flavoprotein (Fp); EF hand domain co ntaining2; ribosomal protein SA; albumin.5. Found the major metabolic pathway that participated by different expre ssed proteins through KEGG pathway analysis. They are RNA degradation p athway; Purine metabolism pathway; Ribosome pathway; Glycolysis/Glucone ogenesis pathway; HIF-1signaling pathway.Gonelusion1.Through the identified in GO annotations different proteins, found tha t these proteins are mainly to achieve its function from biological proc esses such as the development of the nervous system, transcription etc.2.
