Objective:The analysis of the present study was to investigate the current prevalence of atrial fibrillation population in YanBian area, to discuss the distribution of genetic polymorphisms of NPPA in healthy of Korean and Han population in Yanbian area and assess the association between the genetic polymorphisms of NPPA and atrial fibrillation in Korean and Han population, to provide scientific basis for the prevention of atrial fibrillation in YanBian area.Methods:1.The genetic DNA was extracted from136atrial fibrillation patients(58korean and78Han) and300controls (144korean and156Han).2. Genomic DNA was extracted from blood samples of all patients using a TIANamp blood DNA kit according to the standard protocol.3. The single nucleotide polymorphisms of rs2270915、rs5063were genotyped by PCR analysis.4. Specific restrictive endonucleases were used to detect the genotype of r2270915、rs5063in specimens.5. The PCR products were purified for sequencing with sanger method.Results:1.The polymorphism of rs2270915,136AF group AA、AG、GG genotype frwquency werel0%、34%、57%;300control group genotype frequency were15%、49%、37%. significient difference in the distributions of two groups (P=0.000). The distribution of genotypes in AF group and control group had significant difference (P=0.000), The frequency distribution of A and G alleles of two accounted for27%and73%in AF group;39%and61%respectively in the control group.rs2270915frequencies of the three kinds of genotypes (AA、AG、GG) in Korean AF group were9%、38%、52%; control group were12%、56%、30%. The distribution of genotypes in AF group and control group had significant difference (P=0.003), The frequency distribution of A and G alleles of two accounted for29%and71%in AF group;41%and59%respectively in the control group, significant difference in the distributions of A and G allele frequencies was observed between two groups (P=0.007). In Han AF group were10%,29%,60%; control group were12%、52%、36%。The distribution of genotypes in AF group and control group had significant difference (P=0.002), The frequency distribution of A and G alleles of two accounted for25%and75%in AF group;38%and62%respectively in the control group, significant difference in the distributions of A and G allele frequencies was observed between two groups (P=0.006).2. The polymorphism of rs5063,136AF group AA、AG、GG genotype frwquency were32%、52%、18%;300control group genotype frequency were32%、50%、18%. there were no significient difference in the distributions of two groups(P=0.975). The distribution of genotypes in AF group and control group had no significant difference (P=0.970), The frequency distribution of A and G alleles of two accounted for57%and43%in AF group;56%and44%respectively in the control group. rs5063frequencies of the three kinds of genotypes (AA、AG、GG) in Korean AF group were22%、53%、54%; control group were24%、50%、26%. The distribution of genotypes in AF group and control group had no significant difference (P=0.860), The frequency distribution of A and G alleles of two accounted for49%and51%in AF group;48%a nd52%respectively in the control group. There were no significant difference in the distributions of A and G allele frequencies was observed between two groups (P=0.980). In Han AF group were38%、49%、13%; control group were39%、49%、12%。The distribution of genotypes in AF group and control group had significant difference (P=0.990), The frequency distribution of A and G alleles of two accounted for63%and37%in AF group;64%and36%respectively in the control group, significant difference in the distributions of A and G allele frequencies was observed between two groups (P=0.960).3. Logistic regression analysis showed that, rs2270915AG gene(OR=1.165), GG gene(OR=0.930) was independent risk factors for Korean patient with artial fibrillation in Yan Bian area. rs2270915AGgene (OR=0.490), GGgene(OR=1.197), was also independent risk factors for HAN patients with atrial fibrillation in YanBian area. According to the multi-factor logistic analysis in korean, the OR value between AF and coronary disease、 Hypertention、AG、GG genotypes of rs2270915wasl.092、1.270、1.165、0.930respectively. According to the multi-factor logistic analysis in Han, the OR value between AF and coronary disease, hypertension、AG、GG genotypes was2.384、0.53、0.490、1.197respectively.Conclusions:1. The polymorphisms of rs2270915of NPPA was associated with the risk of AF in Korean and Han population of YanBian area. There is no obvious correlation between the rs5063polymorphism with atrial fibrillation Korean and Han population in YanBian area.2. LAD and AG,、 GG genotypes of rs2270915maybe more increase the AF risk in Korean and Han population in YanBian area.3. In YanBian area Korean and Han population rs2270915has no national difference.
