In recent years, with the continuous improvement of people’s living standards, changes in eating habits and diet as well as an aging population, the mortality and morbidity of colorectal cancer are on the rise. In the digestive malignancy, the mortality and morbidity of colorectal cancer are chasing after gastric and esophagus cancers. The majority of patients are in advanced stages when discovered.So far, CEA, CA19-9and CA24-2are recognized as the most credible of serum tumor markers associated with colorectal cancer but with low diagnostic sensitivity and specificity. The aim of this study was to investigate whether the fatty acid binding to albumin is detectably and significantly altered in colorectal cancer patients compared with healthy people and analysis its value in the diagnosis of colorectal cancer.141cases of colorectal cancer patients, who were treated between October2012to March2013, and180healthy volunteers were included in this study. Commercial16Doxyl stearic acid was used as spin probe. The electron paramagnetic resonance spectrometer could get the spectroscopy of stearic acid binding to serum albumin. The five parameters (△H1△H2△H3△H4△H5, relative line width and peak width) which obtained from the spectroscopy could be applied in the establishment of discriminant function by using the discriminant analysis statistics method. The discriminant analysis statistical method was used for getting effective biomedical information from spectroscopy and establishing the discriminant function and exploring the value of colorectal cancer diagnostic serum albumin electronic paramagnetic spectroscopy.The fatty acid binding to albumin in colorectal cancer patients, compared with the healthy volunteers’, was significantly different. We have got five parameters by analyzing the spectroscopy. At the same time, we have got the discriminant function by fisher discriminant analysis: F1=338.904△H1+88328.145△H2-103.109△H3+96423.334△H4+265.178△H5-255051.924; F2=328.375△H1+88226.653△H2-116.781△H3+96324.452△H4+259.369△H5-254166.845.Diagnostic specificity is87.9% and sensitivity is86.1%, respectively. Confirming the screening test has great diagnostic value.The fatty acid binding to albumin was detectably and significantly altered in colorectal cancer patients compared with healthy people. Cancer associated alterations to albumin can be assessed by spin-label EPR. Spin-labeled serum albumin electron paramagnetic resonance spectroscopy in diagnosis of colorectal cancer with high diagnostic sensitivity and specificity. Spin probe for diagnosis of colorectal cancer might be a useful tool and the method has the potential to become a new clinical laboratory method for diagnosis of colorectal cancer.
