Objective:The aim is to offer informations on the genomic and sero-prevalence of Human Bocavirus (HBoV) in healthy Chinese plasma donors, plasma pools, plasma derivatives, and to acquire phylogenetic characteristics of HBoV in screened areas.Methods:Based on the ABI7900HT Fast Real-time PCR System platform, we established a specific and sensitive Real-time PCR(SYBR Green) assay to detect HBoV DNA. On the basis of method constructed above, we tested6096individual plasmas of each month,241plasma pools and326plasma derivatives [including65batches of albumin,197batches of intravenous immunoglobulinfrom (IVIG),47batches of anti-hemophilic factor Ⅷ (FⅧ),12batches of Fibrinogen (fibrinogen, Fg),5batches of Prothrombin Complex Concentrate (PCC)] for the genomic prevalence of HBoV. Positive samples were confirmed by nested-PCR and further amplified for a segment of820bp used for sequence analysis and phylogenetic studies. Anti-HBoV-IgG and IgM were also tested to obtain the status of HBoV antibodies in three plasma groups (Group1, HBoV DNA>104copies/mL; Group2, HBoV DNA>5×102copies/mL but below104copies/mL; Group3, HBoV DNA negative).Results:The genomic prevalence of HBoV in healthy Chinese plasma donors were9.06%, with virus loads of5.01×102-3.02×106copies/mL. Of the241plasma pools,43(17.84%) were HBoV DNA positive, and3confirmed with loads higher than104copies/mL. HBoV DNA was detectable in44of326(13.50%) plasma derivatives, including2(3.08%,2/65) albumin samples,14(7.11%,14/197) IVIG samples,26(55.32%,26/47) anti-hemophilic factor VIII samples,2(16.67%,2/12) Fg samples, while five PCC samples were all HBoV DNA negative. Serological analysis of HBoV antibodies according to virus loads showed a higher prevalence of IgG antibody in HBoV DNA positive group than that in DNA negative group, and a lower prevalence of IgM antibody in the low DNA level group than that in the DNA high level group. Phylogenetic analysis demonstrated genotype1was the prevalent genotype of HBoV in healthy Chinese plasma donors of screened areas.Conclusions:HBoV genotype1were detectable with high prevalence rate in healthy Chinese plasma donors, plasma pools and plasma derivatives at low levels. Further study is needed to determine whether HBoV pose a threaten to blood safty.
