Objective: To study the expression of OCT4and OPN in gastric carcinoma and itscorrelation with clinicopathologic significance.Methods: In34pairs of fresh gastric cancer carcinoma, the mRNA level of OCT4and OPN was determined by real-time PCR. In143cases of invasive gastric carcinoma,the expression of OCT4and OPN proteins were determined by the immunohistochemicalstaining, respectively. The relationship between these proteins and clinicopathologicfeatures in gastric carcinoma was analyzed by χ2test and spearman rank correlationanalysis. The survival analysis was analyzed by Kaplan-Meier’s method.Results:(1) In34pairs of fresh gastric carcinoma, the relative expression level ofOCT4and OPN was higher than that in normal gastric tissue(190.0±8.873vs79.28±7.386),274.44±22.47vs75.72, p<0.05). In145cases of gastric carcinoma, thepositive rates of OCT4and OPN protein were68.3%and64.8%, respectively,(p<0.05).(2) Immunhistobiochemical staining showed more OCT4positive cells in patients withadvanced N/TNM stages, or gastric type gastric caner, or serous involved that in lowerN/TNM stages,or intestinal type gastric caner,or serous not involved (p<0.05).And alsoimmunhistobiochemical staining showed more OPN positive cells in patients withadvanced M/TNM stages, gastric type gastric caner, or serous involved that in lowerM/TNM stages, or intestinal type gastric caner, or serous not involved (p<0.05).(3)Spearman rank correlation analysis shown OPN correlates with OCT4positively both inmRNA and protein level.Conclusion:(1) OCT4and OPN testing could predict the invasive behavior andhigher clinical TNM stages of gastric carcinoma;(2) The over-expression of OCT4and OPN correlates with the metastasis of gastric carcinoma and unfavorable prognosis.
