Objective] The purpose of this study was to investigate the effect of Th17cells and Treg cells in the pathogenesis of periodontitis by the establishment of experimental periodontitis in rats.[Methods]7-8week-old male Sprague-Dawley rats were randomly divided into two groups of12rats each:normal Contral group, periodontitis group. To induce experimental periodontitis model,4-0silk ligature was tied around second maxillary,with sugar water and inoculation of00.4rats each group was executed.,in the second weeks, the fourth weeks,and the sixth weeks after the model of success-Peripheral blood was collected from cardiac sacrifice under10%chloral hydrate general anesthesia,.Gingival tissue was quickly removed to store in liquid nitrogen to detected transcription factors ROR-yt and Foxp3expression by Real Time RT PCR.the left maxillae was stained by methyl blue to measure alveolar bone loss(ABL); the right maxillae was histological examinated with Trap staining and TIE staining;The proportion of Th17cells and Treg cells in peripheral blood was detected by flow cytometry; the amount change of serum IL-6, IL-10, IL-17, TGF-β, IFN-γ was analysised by ELISA. SPSS17.0statistic package was used for statistical analysis of experimental results.[Results]14、28days after modeling,Weight increase of rats in model group was not much as normal group with statistical significance(P<0.05).Probing depth and alveolar bone loss in periodontitis group is more serious in normal group (P <0.05).The proportion of Th17cells and transcription factor ROR-yt, cytokine IL-17, IFN-γ expression was increased gradually with inflammation in periodontitis group(P<0.05); and in the mean time, the proportion of Treg cells and the transcription factor Foxp3was elevated(P<0.05).significant differences of cytokines IL-10was detected in4week and TGF-β were in4and6weeks (P<0.05). The Serum level of IL-6during the all time points was no significant difference compared with each other(P>0.05).[Conclusion] Using ligating floss with sugar water and inoculation of Porphyromonas gingivalis can successfully induce experimental periodontitis modelafter2weeks. Th17cells can mediated inflammation in the pathogenesis of periodontitis, promote bone tissue destruction and resorption. Treg cells can inhibit the process of inflammation in experimental periodontitis rat model...
