ObjectiveIn order to analyze and discuss the expresstion of Thl7/CD4+FOXP3+Treg cells involved in the pathogenesis of childhood asthma, and provide theoretical basis for the prevention and treatment of childhood asthma,we detect Th17 cells,CD4+FOXP3+Treg cells and related cell factors IL-17, IL-10 in peripheral blood of different periods asthma children.Methods1.In this research,we took children with asthma children(n=103) as research subject,and all samples including patients and healthy children were collected the First affiliated hospital Hospital of Zhengzhou University.According to the condition of asthma children, they were divided into two groups:acute attact group(n=71) and alleviated period group(n=32).According to the severity of illness,acute attact asthmatic group was divided into three groups:mild geoup(n=30),moderate group(n=26),severe group(n=15).The healthy children were enrolled as healthy control group(n=30).2.We isolated peripheral blood mononuclear cells(PBMC) and detected the percentage of Th17 cells and CD4+FOXP3+regulatory T cells of PMBC by flow cytometry (FCM).The expression levels of IL-17 and IL-10 in plasma were tested by Enzyme-linked Immuno Sorbent Assay (ELISA).Results1. Th17 cells in PBMC and IL-17:The acute attact asthma group were higher than alleviated period asthma group and those of the healthy control group, alleviated period asthma group was higher than healthy control group (P<0.05). The acute severe group was significantly higher than the moderate asthmatic group and slight group with statistic significance. (P<0.05).2. The levels of CD4+ FOXP3+Tcells and IL-10:The acute asthma group were significantly lower than alleviated period asthma group and healthy control group(P <0.05). Comparing the acute severe group with moderate group and slight group,the acute severe group was significantly decreased.The moderate group was lower than slight group (p<0.05)..3. The percentage of Th17 cells in PBMC were positively correlated with the levels of IL-17 expression levels in plasma(r=0.817,p<0.0001),and Th17 cells/ IL-17 were positively correlated with the course of childhood asthma (r= 0.649, 0.873,P<0.05)4.There was positive correlation between percentage of CD4+FOXP3+regulatory T cells in PBMC and expression levels of IL-10 in plasma(r=0.235,p<0.05).which had negatively correlated with the conditons of childhood asthma (r=—0.599, 0.241, P<0.05).5. The expression levelsof Th17 and Treg cells in asthmatic children were positively related.Conclusionsl.The Thl7 cells in PBMC/IL-17 involved in the pathogenesis of in children asthma as proinflammation factors and increased with the condition of asthmatic children.The CD4+FOXP3+regulatoryT cells in PBMC and associated cytokine IL-10 participated in the pathogenesis of childhood asthma.We speculated on this kind cell may be have fuction defect and IL-10 exists on synthetic dysfunction in moderate and severe childhood asthma patient.2.Th17/CD4+FOXP3+Treg cells imbalance state could be corrected after long-term regular glucocorticoid (budesonide) treatment,and the drug inhibited systemic inflammation in childhood asthma;IL-10 expression level recovery was slow.3.Th17/IL-17 shaft as proinflammation factors involved in the development of childhood asthmatic pathophysiological events;CD4+FOXP3+Treg cells in asthma children PBMC with IL-10 expression levels have positively correlation,pointing to CD4+FOXP3+Treg cell/IL-10 involved in the pathogenesis of childhood asthma and played the inhibitiory action in asthma inflammation process.IL-17 and IL-10 is expected to be used as activity index of asthma in children,and also are expected to provide new targets for asthma treatment.4.Th17/Treg cellular immune imbalances closely related with airway inflammatory response of childhood asthma,which make up for the Th1/Th2 immune cells imbalances defect,and further complemented the pathogenesis of childhood asthma, and opened up new ideas for the treatment of asthma.
