ObjectivesTo investigate the expression of p53in adenoid cystic carcinoma ACC-2cell under the effect of arsenic trioxide(As2O3).MethodsACC-2cells were cultured in vitro. Arsenic trioxide of different concentrations(1.0、2.0、4.0、8.0μmol/L) and RPMI-1640culture solution were applied to cells in logarithmic growth phase for24hours and48hours as treatment groups and control groups. Cell metamorphoses after As2O3induction were observed under inverted phase contrast microscope. Expression changes of p53mRNA and protein were determined by reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry respectively.Results1.The proliferation of ACC-2cells was significantly inhibited by As2O3, which was time-dependent and concentration-dependent.2. Positive responses for anti-p53antibodies were showed in cultured ACC-2cells.The expression of p53antigen was significantly higher in the control group. Meanwhile the expressions in treatment groups decreased gradually with the inhibition of different concentrations of As2O3(2.0,4.0,8.0μmol/L) whereas there is no statistical difference between1.0μmol/L and2.0μmol/L concentrations.3.Point mutation in123th site of p53mRNA were detected in cultured ACC-2cells. The expression of p53mRNA was significantly higher in the control group whereas the expressions in treatment groups decreased gradually with the inhibition of different concentrations of As2O3(1.0,2.0,4.0,8.0μmol/L)ConclusionAs2O3has the inhibitory effect on the expression levels of mutant type p53mRNA and protein on adenoid cystic carcinoma ACC-2cell line.
