Purpose:Analysis the expression of GPC3mRNA in the peripheral nucleated cells of AFP-negative HCC patients. To investigate the clinical value of GPC3mRNA detecting in the diagnosis and therapeutic monitoring of AFP-negative patients with hepatocellular carcinoma.Method:Collected the peripheral blood of69cases of AFP-negative patients with he patocellular carcinoma (25cases of patients with advanced multiple metastases,20cases of locally advanced patients with no metastasis,24cases of patients with early surgery, all patients were pathologically confirmed ashepatocellular carcinoma),38cases of benign liver disease patients,36cases of healthy volunt eers. Extraction of the total RNA, Analysis the expression of GPC3mRNA in t he peripheral nucleated cells of AFP-negative HCC patients. Follow-up study o f patients without distant metastases, Endpoint was six months after treatment, a positive result is the emergence of recurrence. Using statistical methods to a nalyze the relationship between GPC3mRNA expression in AFP-negative patient s with hepatocellular carcinoma and tumor recurrence.Results:1.39cases of nucleated cells GPC3mRNA positive express in69cases of AFP-negative HCC patients (positive rate56.5%),19cases in benign liver lesi ons and healthy volunteers appear GPC3mRNA positive expression in periphera1blood (positive rate25.7%), by the chi-square statistics treatment (P<0.05), the difference is statistically significant.2. Late with metastasis in patients with positive expression rate of84.0%(21/25), locally advanced found no distant m etastases in patients with positive expression rate of40.0%(8/20), early postop erative patients with positive expression of41.7%(10/24), patients with metasta sis positive expression rate and expression level was significantly higher found no obvious shift, the chi-square statistical treatment (P<0.05), the difference was statistically significant.3. The follow-up found no significant shift and per ipheral blood GPC3mRNA express positive patients, compared with GPC3mRN A express negative patients, short transfer time of recurrence, transfer rate incre ased. Conclusion:GPC3mRNA detection of peripheral blood nucleated cells can be used for AFP-negative HCC patients auxiliary diagnosis, therapeutic effect monitoring and prognosis judgement.
