Objective To explore the changes and significance of transient receptor potential canonical6(TRPC6) in pregnant woman with preeclampsia.Methods From August2012to January2013, in this study we recruited20pregnant women with mild preeclampsia,23with severe preeclampsia and selected25pregnant women in Cesarean section with social factors and the head disproportion factors as a control group in the Department of Obstetrics of the Affiliated Hospital Of Qingdao University Medical College. TRPC6of each group was measured by Enzyme-linked immunosorbent assay (ELISA). We analyzed the correlation of TRPC6and blood pressure、Biochemical indexes and24h-urinary proteins in three groups. The mRNA and protein in myometrium and placenta were measured by quantitative real-time PCR and western blot.Results (1) The serum levels of TRPC6in mild preeclampsia group and severe preecla-mpsia group were (265.17±94.333)pg/mL and (288.32±119.449) pg/mL, respectively. Both were significantly higher than the control group (191.20±88.594)pg/mL (F=5.98, q=3.43,q=4.68.P<0.05). There was no significant difference between mild preeclampsia group and severe preeclampsia group(q=1.05,P>0.05).In mild and severe preeclampsia group, serum level of TRPC6were both positively related with systolic and diastolic blood pressure (r=0.712, P<0.05; r=0.774, P<0.05),and obviously related with the24-hour total urinary protein (r=0.841, P<0.05;r=0.897, P<0.05).But the level had no correlation with triglyceride (TG).total cholesterol (TC),high-density lipoprotein (HDL),low-density lipoprotein (LDL)(P>0.05).(2) The TRPC6mRNA expression in placental and myometrial tissues of mild preeclampsia group and severe preeclampsia group were both exceeded that of the control group. There was no obviously difference between the expression of TRPC6mRNA in the mild preeclampsia group and severe preeclampsia group (q=0.79、2.34,P>0.05).(3) In placenta and myometrium tissue the expression of TRPC6protein in the groups of mild preeclampsia and severe preeclampsia were both evidently increased comparing with the control group (placenta:F=10.08. q=5.08, q=9.17, q=3.59, P<.05; myometrium:F=20.10, q=8.75, q=13.59, q=4.39, P<0.05.(4) In placenta tissue the serum level of TRPC6was positively associated with the TRPC6mRNA and protein(r=0.31, r=1.37, P<0.05). In myometrium tissue the serum level of TRPC6was significantly associated with the TRPC6protein(r=1.36, P<0.05), but there is no difference with the TRPC6mRNA(r=0.11, P>0.05).Conclusion The up-regulation of TRPC6mRNA and excess expression of TRPC6protein in placenta and myometrium may induce higher level of TRPC6in serum. There may be some correlations between the TRPC6channel activated by damaged vascular endothelial cell and the proceeding of preeclampsia.
