Objective:To investigate the expressions with clinical significance of high mobility group boxl protein(HMGB1),nuclear-factor-kappaB(NF-κB) and tumor necrosis factor-a(TNF-a)in human gliomas.Methods:The expressions of HMGB1、NF-κB and TNF-a proteins were examined with the S-P immunohistochemical method in52cases of human gliomas and10cases of normal brain tissues.Results:The datas of Immunohistochemistry experimental showed that HMGB1、NF-κB and TNF-a were all expressed in human gliomas.The expressions of HMGB1in different pathological grading of glioma HMGB1had no obvious changes,while the expression of NF-κB was increased.With the deterioration of the tumor, the expression of TNF-a was decreased. Immunohistochemical results showed that the positive expression rate of HMGB1in grade glioma was69.23%, and the expression rate of NF-κB in low gliomas(Ⅰ,Ⅱ) was40.90%, while the rate in high grade glioma was72.41%.The expression rate of TNF-a in low grade gliomas(Ⅰ,Ⅱ) was73.91%, while in high grade gliomas (Ⅲ, Ⅳ) was20.68%。And Spearman correlation analysis showed that there was a significant different between the low-grade gliomas (Grade Ⅰ and Grade Ⅱ)and the high-grade gliomas (GradeⅢ and Grade Ⅳ)in the expressions of TNF-a and NF-κB or HMGB1and NF-κB or HMGB1and TNF-α in human gliomas (P<0.05).Conclusion:1、HMGB1expression exist in different pathological grading of human gliomas, while does not exist in normal brain tissues.It suggested that HMGB1plays a significant role in the emergence and development of human gliomas, but HMGB1has no obvious relationship between degree of differentiation of human gliomas.2、NF-κB protein does not exsit in normal brain tissues,while the expression levels of NF-κB in low grade glioms (Ⅰ/Ⅱ) significantly are lower than those in high grade gliomas(Ⅲ/Ⅳ).It suggested that there is obvisous relationship between NF-κB and the deterioration of the cancer, and the higher level of malignant degree was, the higher the expression of NF-κB will be.3、 TNF-α expression exsit both in normal brain tissues and gliomas, the expression of TNF-a in glioma was negatively related to the pathological grading of glioma.The expression of TNF-a in glioma(Ⅰ/Ⅱ) was significantly higher than in gliomas(Ⅲ/Ⅳ).This showed that with the deterioration of the tumor, TNF-a, instead, was expressed lower level.4、TNF-a may inhibit the proliferation,transfer, attacks of human gliomas, while HMGB1, NF-κB may promote the proliferati, invasion and antiapoptotic of human gliomas.
