Objective:Using a mice model of asthma, to detect the effect of NGF on maturation and phenotype (CD103+DCs) of lung DCs preliminarily.Methods:32balb/c mice were divided into four groups randomly: control group, asthma group (mice were sensitized and challenged with OVA), IgG group (treatment with IgG) and anti-NGF group (treatment with anti-NGF antibody), with8rats in each group.24hours after the last challenge, the expression of NGF in serum and lung tissue were tested,the behavior of mice, airway responsiveness,pathological changes in lungs and IFN-y and IL-4in serum were measured. Maturation (CD80, CD86, MHC-II) and CD103expression of mice lung DCs were investigated.Results:(1) Compared with control group, NGF expression levels were observed to be increased in the lungs and serum of asthmatic mice (P<0.05);there was no significant change between IgG group and Asthma group(P>0.05),while the mice in anti-NGF group performed lower expression levels of NGF(P<0.05); (2) Compared with control group,the asthma symptoms,airway responsiveness,airway inflammation and Th2response(characterized by up-regulation of IL-4and down-regulation of IFN-γ) were observed to be severer in asthmatic mice(P<0.05);there was no significant change between IgG group and Asthma group(P>0.05),while the mice in anti-NGF group performed attenuated asthma symptoms,airway responsiveness,airway inflammation and Th2response;(3) Compared with control group,the expression of DCs surface molecules (CD80,CD86and MHC class II) and the proportion of lung CD103+DCs were increased in asthmatic mice (P<0.05);there was no significant change between IgG group and Asthma group(P>0.05),while NGF neutralization inhibits maturation and the percentage of CD103+DCs;In addition, the proportion of CD103+DCs showed positive correlated with the levels of serum NGF in mice.Conclusions:1.NGF promotes maturation of dendritic cells in the lung of OVA-sensitized asthmatic mice.2.NGF increases the proportion of CD1031DCs in the lung of OVA-sensitized asthmatic mice.
