| Objective:To explore the effect of nerve growth factor(NGF) on the the expression of lung Muc5ac and TH1/TH2 cytokines in OVA-induced asthma rats.Method:27 healthy SD rats were randomly divided into three groups:control group(nï¼8),asthma group(OVA induced,nï¼9) and a NGF group(OVA induced rats treated with NGF,nï¼10).24 hours of treatment,rats were anatomized to measure serum interleukin(IL)-4,interferon(IFN) -γand observe the degree of infiltration of inflammatory cells around bronchus were scored with optical microscope.The express of lung MucSac were determined by immunohistochemical staining.Result:The score of the degree of infiltration of inflammatory cells around bronchus expressed by median were 1 in control group,3 in asthma group and 4 in asthma rat treated with NGF.There were dramatically statistics significance (Hï¼10.03,p<0.05).The expression of IL-4 were 99.2,185.4 and 271.8pg/ml in control group,asthma group and asthma group treated with NGF.There are dramatically statistics significance among three groups(Fï¼9.246,P<0.01).The expression of IFN-γwere 177.2,92.3 and 42.7pg/ml in control group,asthma group and asthma group treated with NGF.There are dramatically statistics significance among three groups(Fï¼17.28,P<0.01).The expression of lung Muc5ac, expressed with positive areas,were 501.75±270.58/um2,1330.22±662.87/um2 and 2033.90±892.28um2 separately in control group,asthma group and asthma group treated with NGF.There are dramatically statistics significance among three groups(Fï¼11.18,P<0.01). Conclusion:The express of lung Muc5ac increase in OVA-induced asthma rat.Nerve growth factor may play an important role in worsen airway inflammation. There were dramatically statistics significance(Hï¼13.75,P<0.05).The level of serum IL-4 were 99.2,185.4 and 262.0pg/ml separately in group A,B and C.There were dramatically statistics significance among three groups(Fï¼7.706, P<0.05).The level of serum IFN-γwere 177.2,91.8 and 120.7pg/ml separately in group A,B and C.There were dramatically statistics significance among three groups(Fï¼6.731,P<0.05).The expression of lung Muc5ac,expressed with positive areas,were 501.75±270.58/um2,1330.22±662.87/um2,715.88±282.49/um2 separately in control group A,B and C.There were dramatically statistics significance among three groups(Fï¼11.82,P<0.01).ConclusionThe expression of lung MucSac increase in OVA-induced asthma rat.Exogenous hydrogen sulfide may play an important role in alleviate airway inflammation and protect from asthma acute break out. |
