ObjectiveTo identify the species of Hunan area in2012NTM clinical isolates by DNA microarray,and to analyze the drug resistance.Methods1.3157clinical strains were collected from the patients with suspected with TB in the year of2012in Hunan province, then were preliminarily identified with p-nitrobenzoic acid/2-thiophene carboxylic acid hydrazide (PNB/TCH) medium, and cultured and preserved with conventional methods.2.271strains which preliminarily identified with PNA/TCH were analyzed again by DNA microarray.3. The strains of NTM were identified to species and were subjected to anti-mycobacterial drug sensitivity tests in order to better understand the antibiotic resistance of NTM.Results1.271NTM strains were preliminarily isolated from3157clinical strains with PNB/TCH, and271strains were again identified to46Mycobacterium tuberculosis(MTB)(16.9%), and225NTM. Therefore,3157cases of tuberculosis in NTM positive samples in Hunan area accounted for225cases (7.1%),2. Of above225clinical NTM strains identified with DNA microarray, the results showed that there were63M.avium (28%),90M.intracellular(40%),9M.fortuitum (4%),54M. turtle or M. abscessus (24%),3M.Gordon (1.33%),3M. scrofulaceum (1.33%), and3M. kansasii (1.33%).3. TB drug sensitivity test by the absolute concentration method demonstrated that most of the NTM show a high degree of resistance to anti-tuberculous drugsConclusions1.Compared with DNA microarray gene chip technique, traditional PNB/TCH growth test identification of NTM have low specificity;2. The infection rates of NTM in Hunan areas is7.1%in the year of2012, there are mainly M.avium, M.intracellular, M.abscessus and tortoise.3.NTM was highly resistant to conventional anti tuberculosis drugs, if it has been identified as NTM, but without tuberculosis drug susceptibility of conventional test.
