Purpose:To examine the expression level of Oct4in breast cancertissue and further investigate its effect on cell invasion of breast cancer.Methed:Western blot analysis was used to detect the protein level ofOct4in27paired breast cancer tissue and adjacent non-tumoral breasttissue. Oct4expression in breast cancer cells was knockdown bylentivirus-mediated shRNA interference technology. The effect of oct4silencing on the cell invasion was determined by wound-healing assay andcell invasion assay. Four EMT markers (E-cadhrin, alpha-catenin, vimentinand N-cadherin) were analyzed by western blot analysis.Result:The results demonstrated that74%(20/27) patients showedelevated Oct4expression in breast cancer tissue compared with that inadjacent non-tumoral breast tissue. Lentivirus-mediated shRNAsignificantly suppressed Oct4expression. Gene silencing of oct4markedlyinhibited invasion of breast cancer cells. Knockdown of oct4in breastcancer cells induced the expression of epithelial markers E-cadherin andα-catenin that was accompanied by a concomitant reduction of mesenchymal marker N-cadherin and vimentin. These finding suggestedthat Oct4expression was upregualted in breast cancer tissue. Genesilencing of oct4inhibited cell invasion of breast cancer through regulatingEMT.
