OBJECTIVE:To detect the expression of CD44s,CD44v3,CD44v6 in human breast cancer cell line MCF-7 cells treated by TNF-α,analyzing the relationship between CD44 expression modulated by TNF-αand study the effect on the invasion of breast cancer cell line MCF-7. METHODS:MCF-7 were treated by TNF-αfor 24h and 48h respectively,and observed of the proliferation by MTT.The mRNA and protein expression of CD44 were analyzed in human breast cancer cell line MCF-7 cells after TNF-αtreatment for 24h by RT-PCR and Western blot assay.The relationship between CD44 expression and TNF-αwas investigated.An in vitro model of cancer cells was made by the Boyden chamber and the artificial matrix membrane.The number of the cells through the artificial matrix membranein every group was counted to compare the change of the invasion capability among the expermental groups.RESULTS:TNF-αtreatment reduced the level of CD44s mRNA and protein by 16%-38%and 5%-47%respectively.Meanwhile TNF-αtreatment increased the level of CD44v3 mRNA and protein by 5%-52% and 2%-79%respectively.At the same time,TNF-αinduced the express of CD44v6 mRNA and protein by 2%-45%and 4%-35%respectively. CONCLUSION:TNF-αmodulate the express of CD44 in human breast cancer cell line MCF-7 cells,affecting in vitro their migration,invasion.
