At the same time, the mechanism of PI3Kp110α, PI3Kp110β and PTEN were studied. Explore the effection and mechanism of PIK3CA and PIK3CB in colorectal canceration. Explore sensitive biomarker in the early prediction of colorectal adenoma and provide valuable significance with the clinical treatment of colorectal adenoma.Materials and Methods:Expressions and correlations of PI3Kp110α, p110β were detected by IHC. Clinical and pathological factors of PI3Kp110α and PI3Kp110β were detected in colorectal adenoma. Proliferation and apotosis was detected by specfic small interfering RNA. Detect the different effects of PI3Kp110α and PI3Kp110β expression. The differences of PI3Kp110α and p110β expressions were detected by real-time PCR and Western blot in the control group, nonsense group and PTEN RNAi group. Detect proliferation of HCT116by CCK-8. Study on apoptosis and cell cycle of PTEN RNAi by flow cytometry. Bcl-2expression was detected by fluorescent antibody technic.ResultsPI3Kp110α and p110β expressions have significant difference in defferent tissues. The expressions of PI3Kp110α and p110β were positively correlated in LGIN. There were no correlations in colorectal carcinoma, HGIN and normal colorectal mucosa between PI3Kp110α and p110β. PI3Kp110α and p110β expression has correlations with Clinical and pathological factors of colorectal adenoma. PIK3CB expressions in PTEN RNAi group were higher compared with the expressions in nonsense group and control group, while Expression of PTEN was decreased. The proliferation rate of HCT116in PTEN RNAi group on48h,72h was higher than the rate in control group. The ratio of S period in PTEN RNAi group of HCT116was higher than the ratio in control group, and the ratio of G0/G1period in PTEN RNAi group of HCT116was lower than the ratio in PTEN RNAi group. The Bcl-2expression in PTEN RNAi group was higher than the expression in control group.ConclusionPI3Kp110α and p110β can be used as biomarkers in colorectal canceration. PI3Kp110α and p110β may play different part roles in the colorectal canceration. Both PI3BKp110α and p110β promoted the differentiation of cancer. PTEN and PI3BKp110β expressions possibly had regulatory relationships. The deficiency of PTEN can raise the expression of PI3BKp110β and activate the PI3K/AKT pathway. PI3BKp110β played an important role in the PI3K/AKT pathaway. We suspected that lower expression of PTEN can improve PI3BKp110β expression, and then activate the PI3K/AKT pathaway.
