Stem cell transplantation is the treatment of organ damage and neurodegenerative diseases researchfocus on domestic and foreign researchers are neural stem cells, embryonic stem cells are explored, eagerto find out the candidate cells transplanted in the field, but because these cells are faced with ethical widerange of issues, sources, immune rejection, in recent years found that bone marrow mesenchymal stem cells(Bone Marrow Mesenchymal Stem Cells, BMSCs) to some extent, make up for this shortfall. BMSCdifferentiation potential, powerful ability to self-replicate and portability exhibit the characteristics of stemcells to become cell transplantation, gene therapy is important candidate cells, bone marrow mesenchymalstem cell transplantation in the treatment of organ damage, neurodegenerative diseases became life sciencesand medicine research focus.Purpose: The study aims the affinity of bone marreow mesenchymal stem cells (BMSCs) to varioustissues and the possibility for therapeutic transplantation to cure neurodegenerative diseases with BMSCs.Methods: The hippocampal slices, liver slices and lung slices were co-cultured with BMSCs toinvestigate BMSC’s affinity to the tissues above, and immunohistochemistry (NeuN and GFAP) werecarried out to understand the differentiation of BMSCs into neurons and glial cells under microenvironmentof hippocampal slices, liver slices and lung slices.Results:①BMSCs have a highest affinity with hippocampal slices in contrast with the slices of liverand lung.②The BMSCs’ temporal affinity on hippocampal slice was observed as well. At P7, BMSCs hadhighest affinity with hippocampus, but there is a time-related decrease afterward.③Differentiation ofBMSCs occured on hippocampal slices of both WT and Tg mouse. Under the induction of hippocampalmicroenvironment, BMSCs could differentiate into nervous cells and glial cells, but there is no significancebetween WT and Tg mouse.Conclusion: BMSCs may provide an opportunity for the therapeutic transplantation to cure theneurodegenerative diseases and brain injuries. The time-related decrease of cell adhesion to hippocampus shows that the early stem cell transplantation is necessary.
