Objective:1. Research the feasibility of BMMSCs transplanted to the PGCL scaffold to successfully construct the BMMSCs-PGCL patch.2. Observe the growth of the BMMSCs-PGCL patch in vitro in normal myocardium and infarcted myocardium.Methods:BMMSCs in female Sprague-Dawley (SD) rats(weight250g-300g) were separated using adherent separation and selection method, cultured in vitro, and selected3rd generation labeled with DAPI. Production of the BMMSCs(2×1O6/cm2) suspensions were planted on PGCL scaffolds to form the BMMSCs-PGCL patch. After culturing for48hours, observed under electron microscope,and made H&E staining, observed under light microscope.SD rats (weight250g-300g), and ligate left anterior descending coronary artery to make rat model of myocardial infarction (MI). Put BMMSCs-PGCL patch into the normal and infarcted myocardium, and cultured for4weeks. After4weeks, the specimens were taken the detection of pathology to determine the BMMSCs survival on the BMMSCs-PGCL patch.Result:1. Primary culture results:The purified BMMSCs showed the morphology of firoblasts and proliferated in culture medium.2. Respectively under the light microscope and electron microscope, the observation results of BMMSCs-PGCL patch in vitro:it showed that BMMSCs grow three-dimensionally on patch, and the cells and patch were adhesive well.3. The observation results of BMMSCs-PGCL patch transplanted into the normal myocardium:after transplanting the patch one week and five weeks, the section observed contrastly under the laser confocal microscope showed:compared with the first week, there were BMMSCs marked by DAPI in the myocardium of the fifth week.4. The observation results of BMMSCs-PGCL patch transplanted into the infracted myocardium:under the laser confocal microscope showed that there were BMMSCs marked by DAPI in the infracted area. The picture of H&E staining showed that there were BMMSCs in the infracted area.Conclusion:1. BMMSCs can grow on PGCL and be adhesive well.2. The compatibility between PGCL and myocardial tissue is good.3. The complexes of PGCL and BMMSCs can be used for reparation of cardiac tissue engineering...
