Objective: To study the expression of Wnt10b in hepatocellular carcinomatissues (HCC) and para-carcinoma tissues, as well as their correlation withclinicopathological parameters of hepatocellular carcinoma, To discuss thesignificance of its occurrence and development in primary hepatocellularcarcinoma.Methods: Wnt10b mRNA and protein were respectively detected byimmunohistochemistry、real-time fluorescent quantitative PCR and western blotin33cases of hepatocellular carcinoma and para-carcinoma tissues. SPSS17.0statistical software is used for statistical analysis.Results:1)Immunohistochemical results showed: in the33cases of tissues,the expressionof Wnt10b in the HCC tissues was higher than the corresponding adjacenttissues in the22cases of tissues,Wnt10b was highly expressed in HCC tissueand lowly expressed in adjacent normal group,which have significantdifferences (P <0.05);2) RT-PCR and Western blot showed: expression levelsof Wnt10b mRNA in HCC was higher than adjacent tissues, and proteinexpression levels of Wnt10b in HCC was higher than adjacent tissues, the expression were significantly different(P<0.05), Wnt10b gene and proteinexpression trends were showing consistency, Wnt10b mRNA and proteinexpression levels were not related with the patient’s gender, age, tumordifferentiation and clinical stage (P>0.05).Conclusions:1) Wnt10b was as animportant member of the Wnt classical signal pathway, and up-regulatedexpression in HCC, Wnt10b combined with Wnt signal pathway suggested thatWnt signal pathway was one of the classic signal pathways in the developmentof HCC, abnormal activation of Wnt10b may be involved in initiating anddeveloping process of HCC;2) Wnt10b gene and protein expression wereshowing a trend of consistency in the results, the highly expression of Wnt10bmay be related with the level of gene transcription, Wnt10b may plays therole of cancer gene in the occur and development of HCC,Which positivelypromoted the transformation between cancer cells and normal liver cells, ratherthan inhibited, thereby to achieve its biological significance.3) Wnt10b mRNAand protein expression levels were not related with the patient’s gender, age,tumor differentiation and clinical stage, suggested that Wnt10b is not anabsolutely independent factors for liver cancer, the occurrence and developmentof liver cancer is participation of a series of genes and proteins, there are avariety of abnormal pathological process of signal transduction pathways,according to the transmission mechanism of Wnt signal pathway, Wnt10bplayed an important role in the start-up phase of conduction process of the Wntsignal pathway, the abnormally high expression of Wnt10b need to influencetumor development by activating a series of Wnt10b relevant regulatory factorsand downstream target genes,so it is very important to further study the mechanism of Wnt10b downstream target genes, to ascertain the whole Wntsignal pathway mediated Wnt10b in all aspects, and to find and block the signaltransduction of key target and inhibit the proliferation of tumor cell.
